Enzyme-assisted extraction of antioxidants from banana peel

Fruit and vegetable processing wastes contain bioactive compounds such as polyphenols. In this study, polyphenolics were extracted from dried local Anamur banana peels using enzyme-assisted extraction (EAE). Ultrasound-assisted extraction (UAE) with aqueous ethanol and EAE-UAE combination were also applied for comparison. Pectinex ultra SP-L and celluclast 1.5 L in sodium acetate buffer (pH 5) were used for EAE. According to the Box-Behnken design optimization results, the highest total phenolic content (TPC) was obtained by EAE with 1% pectinex ultra SP-L and 0.736% celluclast 1.5 L for 1 h extraction at 50°C with a solid solvent ratio of 1/10 (w/v). The extraction effectiveness was found as EAE (with buffer: 1.746±0.078 mg GAE/mL extract; with water: 1.633±0.014 mg GAE/mL extract) > EAE-UAE combination (1.537±0.020 mg GAE/mL extract) > UAE with 50% aqueous ethanol (0.759±0.008 mg GAE/mL extract). For antioxidant capacity (AC) estimations, FRAP, CUPRAC, DPPH• and ABTS•+ assays were performed. No significant differences were detected among FRAP and CUPRAC ACs of extracts obtained by EAE with buffer or water and EAE-UAE. According to DPPH• assay maximum scavenging capacity (SCmax), the ACs of EAE with buffer and water were found as same, but UAE with 50% aqueous ethanol and EAE-UAE provided higher ACs. DPPH• half maximal effective antioxidant concentration (EC50) values showed that EAE with buffer or water and EAE-UAE combination gave similar EC50 values. According to ABTS•+ SCmax values, no significant differences were observed ACs for EAE with buffer, EAE with water and EAE-UAE. When ABTS•+ EC50 values were compared, similar effectiveness was obtained for EAE with buffer and EAE-UAE. CUPRAC and ABTS•+ assay can be used for comparison of ACs.
