MODELING AND CHARACTERIZATION OF FLUID, MACROMOLECULE, AND NANOPARTICLE TRANSPORT IN FIBROUS COLLAGEN HYDROGELS

The transport of therapeutic agents across body and delivery to tumor tissue is a complex and multiscale phenomena. Although delivery efficiencies have improved over the last two decades, we still lack successful delivery to tumor-microenvironment. A modelling framework that considers physiology of both transport media and transported agents is required to investigate the reasons of the insufficient delivery and achieve the extrapolation from in vitro to in vivo, as well as from animal models to human participants. In order to address this problem, this thesis focuses on modeling and characterization of fluid and species transport in tumor-microenvironment by adapting different computational and experimental methodologies to analyze the transport of therapeutic agents across scales.
