Exploring novel A-I RNA editing sites and ADAR activity in breast cancer

Adenosine to inosine RNA editing is a crucial modification within the epi-transcriptome. It is orchestrated by adenosine deaminase (ADAR) enzymes. This post-transcriptional modification significantly influences protein levels and functions and, hence, is pivotal in normal and cancer cells. However, the functional importance of RNA editing events is under-detected and under-valued, primarily due to technical detection limitations. Here, the combination of in-silico editing site discovery in the 3'UTRs with an experimental pipeline has been adopted to confirm and mechanistically decipher the effect of editing sites on candidate transcripts in breast cancer cells. In addition, our study further investigates the possible interaction of ADAR1 with other RNA-binding proteins to light up their collaborative role in facilitating the 3'UTR editing events. In summary, this study aims to improve our understanding of RNA editing events concerning gene deregulation through epi-transcriptomic mechanisms in breast cancers.
