Hydrogels with hydrophilic polymeric structures which enables to absorb large amounts of water and swell and contract in order to provide with controlled release. In this study, the drug delivery system was composed of diatoms as inner compartment encapsulated by pullulan microspheres and the system was formulated in order to investigate the release profiles of luteolin and doxycycline from diatom incorporated pullulan microspheres in simulated gastrointestinal fluid (sGIF) separately with respect to controlled delivery of orally administered drugs. Luteolin as an anti-inflammatory and doxycycline as an antibacterial drug were used as model drugs for the delivery from diatom incorporated pullulan microspheres separately. The formulation and its suitability for oral delivery were assessed considering the release profiles in the conditions of sGIF including simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) and material properties. Diatoms with loading capacity of 9.78 ±3 % and 10.47 ± 5.4% for luteolin and doxycycline exhibited high loading. The release patterns were examined in terms of drug characteristics and the material properties, which indicated that sustained release patterns were observed in diatoms and the formulated system.
