Optimization of mature embryo based regeneration and genetic transformation of Turkish wheat cultivars
Battal, Abdülhamit; Yücel, Ayşe Meral; Öktem, Hüseyin Avni; Department of Biotechnology (2010)
The objective of this study was to optimize tissue culture, transformation and regeneration parameters of mature embryo based culture of Triticum durum cv. Mirzabey 2000 and Triticum aestivum cv. Yüreğir 89. The effects of auxin type of hormone at different concentrations and dark incubation periods on regeneration capacity were evaluated. Two different hormone types 2,4- dichlorophenoxyacetic acid and picloram were used at three different concentrations 2, 4 and 8 mg/l. Mature embryo derived calli were inc...
Optimization of tissue culture, generation and agrobacterium mediated transformation parameters in winter wheat cultivars (Kızıltan-91 and Bezostaja-01)
Kavas, Musa; Yücel, Ayşe Meral; Department of Biology (2005)
The objective of this study was to optimize tissue culture and regeneration parameters of immature inflorescence culture of Triticum aestivum cv. Bezostaja- 01 and Triticum durum cv. Kızıltan-91. The effects of callus age and vernalisation time of explants on regeneration success were evaluated. For determination of optimum vernalisation time of immature inflorescence, plants subjected to 4°C for 1, 2, 3, 4, and 5 weeks, respectively. Tillers containing immature inflorescences were collected at the same tim...
Optimization of supercritical CO2 extraction of Thymbra spicata oil
Sonsuzer, S; Şahin, Serpil; Yılmaz, Levent (Elsevier BV, 2004-07-01)
The supercritical fluid extraction (SFE) of aroma compounds from Thymbra spicata was studied. Response surface methodology (RSM) was used to optimize the parameters of the supercritical fluid extraction. Independent variables were temperature (40, 50 and 60 degreesC), pressure (80, 100 and 120 bar) and time (30, 60 and 90 min). Dependent variables were yield, monoterpene, sesquiterpene and oxygenated monoterpene contents.
Optimization of ethanol-water extraction of lignans from flaxseed
Zhang, Zhen-Shan; Li, Dong; Wang, Li-Jun; Özkan, Necati; Chen, Xiao Dong; Mao, Zhi-Huai; Yang, Hong-Zhi (2007-10-01)
The optimal conditions for the extraction of lignans from flaxseed were determined. A second-order regression for rotation-orthogonal composite design was used to investigate the effects of three independent variables, namely ethanol concentration (%), extraction temperature (degrees C, and extraction time (h) on the response, the acquired ratio of lignans. The independent variables were coded at five levels and their actual values selected on the basis of results of single-factor experiment. The second-ord...
Optimization of solid-liquid extraction of resveratrol and other phenolic compounds from milled grape canes (Vitis vinifera)
Karacabey, Erkan; Mazza, Giuseppe (American Chemical Society (ACS), 2008-08-13)
Optimization of the solid-liquid extraction conditions for trans-resveratrol, trans-E-viniferin, ferulic acid, and total phenolics from milled grape canes has been investigated. The temperature and ethanol concentration were found to be major process variables for all responses, whereas the solvent to solid ratio was found not to be significant for any of the responses studied. The yields of transresveratrol, trans-epsilon-viniferin, and total phenolics increased with increasing temperature. Maximum yields ...
