Purification of microbially produced ca-l(+)-lactate with a novel technique: drying induced elution of impurities
Özçelik, Damla; Hamamcı, Haluk; Department of Food Engineering (2016)
For the production of calcium L-(+)-lactate (Ca-L(+)-Lactate) fermentation was performed by using Lactobacillus casei. Production with inexpensive raw materials and low cost purification of lactic acid (LA) as Ca-L(+)-Lactate was aimed because LA production from renewable materials is limited due to high production and purification costs. Fermentation conditions were 37°C for temperature, pH was kept at 5.5 by adding CaCO3 as neutralizer and stirring was adjusted at 120 rpm. Fermentation medium consisted of...
Purification and characterization of two forms of endo-beta-1,4-mannanase from a thermotolerant fungus, Aspergillus fumigatus IMI 385708 (formerly Thermomyces lanuginosus IMI 158749)
Puchart, V; Vrsanska, M; Svoboda, P; Pohl, J; Ögel, Zümrüt Begüm; Biely, P (Elsevier BV, 2004-11-01)
Two extracellular endo-β-1,4-mannanases, MAN I (major form) and MAN II (minor form), were purified to electrophoretic homogeneity from a locust bean gum-spent culture fluid of Aspergillus fumigatus IMI 385708 (formerly Thermomyces lanuginosus IMI 158749). Molecular weights of MAN I and MAN II estimated by SDS-PAGE were 60 and 63 kDa, respectively. IEF afforded several glycoprotein bands with pI values in the range of 4.9–5.2 for MAN I and 4.75–4.9 for MAN II, each exhibiting enzyme activity. MAN I as well a...
PURIFICATION AND CHARACTERIZATION OF HUMAN MENINGIOMA M2-TYPE PYRUVATE-KINASE
MELLATI, AA; YUCEL, M; ALTINORS, N; Gündüz, Ufuk (1993-10-01)
The M2-type pyruvate kinase was purified from human meningioma by ammonium sulfate precipitation, followed by ion exchange and affinity chromatography. The specific activity of the purified enzyme was 33.4 U/mg with a yield of 6.5%.
