Mammary gland is a dynamic organ that undergoes dramatic changes in response to age, circulating hormones and reproductive status. The extensive growth, regeneration and remodelling abilities of the mammary gland is of specific interest to better understand the molecular mechanisms deregulated in breast cancers. To this end, we hypothesized alternative polyadenylation (APA) as a mechanism that generates mRNA isoforms with different lengths, to be dynamically regulated at different stages of mammary development. APA is known to be regulated in a tissue and developmental stage specific manner and is linked to mRNA 3’UTR isoform variability, which may result in altered protein levels and/or functions. We used a combinatorial in silico and in vitro approach and delineated a dynamic pattern of APA in gestation, lactation and involution stages. Experimental confirmation of 3’UTR isoforms indeed provided preliminary data for future studies to provide a possible link between mammary tissue development, APA and breast cancer.

Dynamic transcriptional events mediated by estrogen receptor alpha
Ayaz, Gamze; Yasar, Pelin; Olgun, Çağla Ece; Karakaya, Burcu; Kars, Gizem; Razizadeh, Negin; Yavuzl, Kerim; Turan, Gizem; Muyan, Mesut (Frontiers in Bioscience, 2019-01-01)
17beta-estradiol (E2), the main circulating estrogen hormone, is involved in a wide variety of physiological functions ranging from the development to the maintenance of many tissues and organs. The effects of E2 on cells are primarily conveyed by the transcription factors, estrogen receptor (ER) alpha and beta. The regulation of responsive genes by the well-defined ER alpha in response to E2 relies on complex and highly organized processes that dynamically integrate functions of many transcription regulato...
Structural analysis of a 50 kb region on 17q23
Akman, Begüm H; Erson Bensan, Ayşe Elif; Department of Biology (2007)
17q23 amplicon is one of the many chromosomal regions that undergo amplification in breast tumors. Such amplicons harbor proto-oncogenes that may be overexpressed due to gene amplifications. Copy number analysis in breast cancer cell lines and breast tumors identified several independently amplified regions within the 17q23 amplicon, suggesting that a number of genes are selected for amplification as they may independently contribute to tumor formation and progression. To characterize distinct amplicons on ...
Multidrug resistance in logally advanced breast cancer
Atalay, Mustafa Can; Gündüz, Ufuk; Department of Biotechnology (2004)
Breast cancer is the most frequently detected cancer among women. Early diagnosis leads to long term survival when the patients are treated with surgery, radiotherapy, chemotherapy, and hormone therapy. Unfortunately, advanced disease could still be encountered in some patients resulting in a poorer prognosis. The primary treatment modality is chemotherapy for this group of patients. Drug resistance is a serious problem resulting in the use of different drugs during chemotherapy and knowing the possibility ...
Meme kanseri tedavisine yönelik işlevleri kontrol edilebilir yapay transkripsiyon baskılayıcıların geliştirilmesi
Özen, Can; Muyan, Mesut; User, Damla Sırma; Yaşar, Pelin(2016)
Novel BRCA2 pathogenic genotype and breast cancer phenotype discordance in monozygotic triplets
Duzkale, Neslihan; EYERCİ, NİLNUR; Oksuzoglu, Berna; Teker, Taner; Kandemir, Olcay (Elsevier BV, 2020-04-01)
BRCA1/2 genes with high-penetrance are tumor suppressor and tumor susceptibility genes that play important roles in the homologous recombination mechanism in DNA repair and increase breast cancer risk. Variants in BRCA1 or BRCA2 are the main causes of familial and early-onset breast cancer. This study investigated pathogenic variant belonging to the BRCA2 gene splice region in monozygotic triplets. A 44-year-old woman was diagnosed with breast cancer when she was 32 years old. Her monozygotic sister had a h...
