Extension of flower longevity in transgenic plants via antisense blockage of ethylene bıosynthesis
Deçani Yol, Betul; Öktem, Hüseyin Avni; Department of Biotechnology (2004)
Ethylene (C2H4) is a very simple molecule, a gas, and has numerous effects on the growth, development and storage life of many fruits, vegetables and ornamental crops. In higher plants, ethylene is produced from L-methionine in essentially all tissues and ACC Synthase and ACC Oxidase are the two key enzymes in the biosynthesis of ethylene. The objective of the present study was to transform tobacco (Nicotiana tabacum L. cv. Samsun) plant with partial sequence of torenia acc oxidase gene in antisense and sen...
ANALYSIS OF PLASMIDS MEDIATING ANT (2'') AND AAC(3)-II BASED GENTAMICIN RESISTANCE IN CLINICAL ISOLATES OF ENTEROBACTERIACEAE
Kocabıyık, Semra (Elsevier BV, 1989-07-01)
The genetics of ANT(2″) and AAC(3)-II mediated gentamicin resistance in Gram-negative bacteria obtained from Hacettepe University Hospital in Turkey, was studied. The plasmid DNA analyzed by agarose gel electrophoresis revealed a heterogeneous plasmid population. Conjugal transfer appeared to be an efficient mechanism for the dissemination of resistance to gentamicin/tobramycin/kanamycin/streptomycin/chloramphenicol/cephalothin, which was the common resistance pattern in the population. Plasmids of 46, 60, ...
Optimization of regeneration and agrobacterium mediated transformation of sugar beet (Beta vulgaris L.)
Baloğlu, Mehmet Cengiz; Yücel, Ayşe Meral; Department of Biology (2005)
In this study, optimization of a transformation and regeneration system via indirect and direct organogenesis in cotyledon, hypocotyl, petiole, leaf and shoot base tissues of sugar beet (Beta vulgaris L. cv. ELK 345 and 1195) was investigated. Two different germination, three different callus induction and shoot induction medium was used for indirect organogenesis of sugar beet cultivar ELK 345. Except cotyledon, other explants (hypocotyl, petiole and leaf) produced callus. However no shoot development was ...
Development of salt resistant transgenic plants by using TaNHX1 and TaSTR genes
Kavas, Musa; Yücel, Ayşe Meral; Öktem, Hüseyin Avni; Department of Biology (2011)
Soil salinity negatively affects agricultural production in Turkey by decreasing the yield and quality. Direct introduction of stress related genes by genetic engineering is one of the most rapid approaches to develop stress tolerant crops. In this study, TaNHX1 gene was isolated from bread wheat and three different local wheat cultivars were transformed with overexpression vectors containing TaNHX1 gene by using Agrobacterium-mediated and particle bombardment gene transfer techniques. Immature embryo and i...
