Four Staphylococcus isolates having both multidrug- and multimetal-resistant ability were isolated from surface water. Further identification of the isolates was obtained through biochemical tests and 16S rDNA gene sequencing. One methicillin-resistant and two methicilin-sensitive isolates were determined as Staphylococcus aureus. The other isolate was identified as Staphylococcus warneri. The antibiotic and heavy metal resistance profiles of the Staphylococcus isolates were determined by using 26 antibiotics and 17 heavy metals. S. aureus isolates displayed resistance to most of the beta-lactam antibiotics tested. All Staphylococcus isolates were resistant to heavy metals including silver, lithium, and barium. Due to a possible health risk of these pathogenic bacteria, a need exists for an accurate assessment of their acquired resistance to multiple drugs and metals.

Purification and characterization of cytoplasmic and proteasome associated chymotrypsin-like proteases from Thermoplasma volcanium
Özdemir, F. İnci; Kocabıyık, Semra; Department of Biology (2003)
In this study, two novel cytoplasmic serine proteases were isolated and characterized from thermophilic archaea Thermoplasma volcanium. The first protease was purified by ion exchange and affinity chromatographies and identified as a chymotrypsin-like serine protease mainly based on its substrate profile and inhibition pattern. The presence of protease activity was analyzed by gelatin zymography which was detected as a single band (35 kDa). Optimum temperature was found to be 60oC for azocasein hydrolysis a...
Waterborne parasitic protozoa removal capacities of wastewater treatment plants with varying processes
Onursal, Aslı; İçgen, Bülent; Department of Environmental Engineering (2022-8-26)
The parasitic protozoa Giardia intestinalis, Entamoeba histolytica, Blastocystis hominis, and Cryptosporidium parvum are causative agents for human giardiasis, amebiasis, blastocytosis (formerly known as hominis infection) and cryptosporidiosis, respectively. These infections are mostly associated with waterborne diseases. Due to the lack of regulations for monitoring these protozoa in the discharge point of wastewater treatment plants (WWTPs), the discharges that reach to surface waters lead to waterborne ...
Intracellular alkaline proteases produced by thermoacidophiles: detection of protease heterogeneity by gelatin zymography and polymerase chain reaction (PCR)
Kocabıyık, Semra (2002-08-01)
In this study 24 thermoacidophilic archeal and bacterial strains isolated from hot-springs and hot-soils were screened for their ability to produce intracellular alkaline proteases. The protease activities of the strains, based on azocasein hydrolysis, showed a variation from 0.6 to 5.1 U. The cell extracts of three most potent producers were further examined and it was found that their proteases exhibited maximum activity at 60-70 degreesC and showed a pH Optimum over a range of pH 7.0-8.5. Gelatin zymogra...
Heavy metal biosorption by white-rot fungi
Yetiş, Ülkü; Özcengiz, Gülay; Dilek, Filiz Bengü (1998-06-26)
In this study, heavy metal biosorption potentials of two white-rot fungi, Polyporous versicolor and Phanarochaete chrysosporium, which are commonly used in wastewater treatment were determined. Biosorption studies were performed for Cu(II), Cr(III), Cd(II), Ni(II) and Pb(II) at the same operational conditions and the effectiveness of both fungi at removing these heavy metals was compared. It was found that both P. versicolor and P. chrysosporium were the most effective in removing Pb(II) from aqeous solutio...
Design rules for suspending concentrated mixtures of solids in stirred tanks
Ayrancı Tansık, İnci (2011-10-01)
This study examines complete off-bottom suspension conditions for slurries containing mixtures of solids at a wide range of concentrations. Binary combinations of five different particles; bronze, two sizes of glass beads, ion exchange resin, and nickel were tested with two impellers: the Lightnin A310 and the down pumping 45 degrees PBT. The effect of particle size ratio and density ratio of the two solid phases on N(js) was investigated. Solids loadings were varied from 3 to 56 wt% (weight percent) with w...
