Inflorescence explants of two winter wheat cultivars, Triticum durum cv. Kiziltan-91 and T. aestivum cv. Bezostaja-01, were used to evaluate the effects of vernalization period of donor plants, callus age and medium composition on regeneration capacity. Donor plants were grown for 7 d and they were exposed to 4 degrees C for 1, 2, 3, 4, and 5 weeks. The maximum inflorescence formation was observed as 79 % at 4 weeks and 73 % at 5 weeks of vernalization period for Kiziltan-91 and Bezostaja-01, respectively. Among 6 different callus induction and regeneration mediums, I-1-R-1 and I-3-R-3 have to be the best responding mediums for Kiziltan-91 and Bezostaja-01, respectively. In Kiziltan-91, calli induced from donor plants, vernalized for 3 weeks, showed a significantly lower regeneration capacity than counterparts vernalized for 4 and 5 weeks. The highest regeneration capacity of 69 % was obtained from 6-week-old calli produced from 4 weeks vernalized Kiziltan-91 donor plants. In contrast to Kiziltan-91 cultures, the effects of vernalization period and callus age on regeneration capacity were not significant in Bezostaja-01 cultures. The maximum numbers of tillers were obtained from 6-and 15-week-old calli for Bezostaja-01 and Kizilitan-91, respectively. In contrast to vernalization period of donor plants, callus age had no effect on seed number.

Genetic composition of four marginally located Anatolian black pine (Pinus nigra subsp. pallasiana) populations determined by SSR markers
Aygün, Sıla; Kaya, Zeki; Department of Biology (2018)
Anatolian black pine (Pinus nigra Arnold subsp. pallasiana (Lamb.) Holmboe) is one of the most economically and ecologically important coniferous tree species of Turkey. Global climate change is obviously going to affect distribution and development of forest tree species and the marginal populations will be the most vulnerable ones. Since these populations are located in outer borders of their natural distribution area, they are expected to have original genetic makeup shaped by unsuitable living condition...
Changes in total protein profiles of barley cultivars in response to toxic boron concentration
Mahboobi, H; Yucel, M; Öktem, Hüseyin Avni (2000-01-01)
In this study, ten-day-old seedlings of barley {Hordeum vulgare L. cultivar Anadolu [boron (B)-tolerant] and Hamidiye (B-sensitive)} were used. Boron-treated plants were grown on H3BO3 solution (final concentration of 10 mM) for five days. Control plants received no B treatment during this period. Total protein patterns were obtained by analysis of total protein extract from root and leaf tissues of control and B-treated plants using two-dimensional gel electrophoresis followed by silver staining. The prote...
Effect of different wheat varieties on pasta quality
Bozkurt, Murat; Mert, Behiç; Department of Food Engineering (2012)
There are about 40 durum wheat varieties registered in Turkey. Around 10 varieties are being commonly cultivated. Amanos 97, Çeşit 1252, Ege 88, Fırat 93, Fuatbey 2000, Burgos, Kızıltan 91, Sarıçanak 98, Svevo and Zenith durum wheat varieties were used in this study. Semolina and pasta have been produced in an industrial plant under constant process conditions from these varieties. In general, pasta quality is determined by three main factors, the raw materials, the production recipe and the production proc...
The utility of NBS-profiling for characterization of yellow rust resistance in an F6 durum wheat population
Tufan, Hale A.; Taşkin, Belgin Göçmen; Maccormack, Ruth; Boyd, Lesley A.; Kaya, Zeki; Türet, M. (Indian Academy of Sciences, 2019-11-01)
Seedling and adult plant (field) resistance to yellow rust in the durum wheat (Triticum turgidum ssp. durum) cross Kunduru-1149 x Cham-1 was characterized using a functionally-targeted DNA marker system, NBS-profiling. Chi-squared analysis indicated a four gene model conferring seedling yellow rust resistance against Puccinia striiformis f. sp. tritici isolate WYR85/22 (virulent on Yr2, Yr6, Yr7 and Yr9). Interval mapping located two QTL for yellow rust resistance on the long arm of chromosome 1B, while Kru...
Transformation of potato with MYB4 transcription factor and evaluation of abiotic stress tolerance and gene expression profiles in transgenic plants
Kalemtaş, Gülsüm; Öktem, Hüseyin Avni; Department of Biology (2011)
Potato (Solanum tuberosum L. cv. Kennebec) was transformed via Agrobacterium tumefaciens (EHA105) harbouring two different binary vectors containing Oryza sativa myb4 gene, which encodes MYB4 transcription factor; under the control of CaMV35S promoter or cold inducible COR15a promoter. The transgenic plants were not growth retarded and there was no significant difference (p<0.05) in their tuber yield compared to wild-type plants. Wild-type and transgenic plants were subjected to abiotic stresses to compare ...
