Pretreatment of poultry litter improves Bacillus thuringiensis-based biopesticides production

2010-04-01
Pretreated poultry litter was used in batch cultures for the production of Bacillus thuringiensis (Bt)-based biopesticide of lepidoptera- and diptera-specific Cry1 and Cry2, diptera-specific Cry4Ba and Cry11Aa and coleoptera-specific Cry3Aa toxins by Bt subsp. kurstaki 81, subsp. israelensis HD500 and subsp. tenebrionis 3203, respectively. Bt kurstaki 81 showed improved growth and produced more toxin in this medium as compared to other subspecies. Base and acid hydrolysis were tested as the methods of substrate Pretreatment. The use of poultry litter pretreated with 2 N HCl yielded 94% more bioinsecticidal protein than 2 N NaOH-pretreated poultry litter when Bt kurstaki 81 was cultured. With appropriate pretreatment, poultry litter demonstrated potential as a valuable raw material for a low-cost complex medium to produce Bt-based biopesticides.

Proteome-wide analysis of the role of expression of bacilysin operon on idiophase physiology of B. Subtilis
Demir, Mustafa; Özcengiz, Gülay; Department of Biology (2013)
The members of the genus Bacillus produce a wide variety of secondary metabolites with antimetabolic and pharmacological activities. These metabolites are mostly small peptides and have unusual components and chemical bonds. These metabolites are synthesized nonribosomally by multifunctional enzyme complexes called peptide synthetases. One of those small peptides, bacilysin, is a dipeptide antibiotic composed of L-alanine and L-anticapsin which is produced and excreted by certain strains of Bacillus subtili...
Removal of chloridazon herbicide from wastewaters using FE/H2O2, UV/ H2O2 and UV/FE/ H2O2

Chloridazon (5 -amino-4- chloro-2-phenyl-3(2H)-pyridazinon), also named as Pyrazon and classified as organochlorine pesticides, is widely used during sugar beets cultivation. Chloridazon (CLZ) being a pesticide with a high solubility in water is likely to end up in surface and groundwater bodies because of its high mobility in soil. Due to its toxic properties, it may cause serious problems on human health and ecological cycle. In the present study, the removal of CLZ herbicide from water was investigated b...
Utilization of pretreated molasses for serine alkaline protease production with recombinant Bacillus species
Calik, G; Pehlivan, N; Kalender, N; Ozdamar, TH; Çalık, Pınar (Informa UK Limited, 2003-05-01)
Recombinant Bacillus amyloliquefaciens, Bacillus cereus, Bacillus subtilis , and Bacillus licheniformis were used for the production of serine alkaline protease (SAP) utilizing chemically and/or physically pretreated molasses. The highest enzyme activity was obtained with r- Bacillus subtilis , with the complex medium involving physically treated molasses having 20 kg m(-3) initial sucrose concentration in small-scale, agitation- and heating rate-controlled bioreactors at t=63 h. Effects of oxygen transfer ...

The members of the genus Bacillus produce a wide variety of secondary metabolites with antimetabolic and pharmacological activities. Most of these metabolites are small peptides that have unusual components and chemical bonds and synthesized nonribosomally by multifunctional enzyme complexes called peptide synthetases. Bacilysin, being produced and excreted by certain strains of Bacillus subtilis, is one of the simplest peptide antibiotics known. It is a dipeptide with an N-terminal Lalanine and an unusual ...
Identification and characterization of hydrolytic enzymes from the midgut of Sunn Pest of wheat (Eurygaster integriceps)
Ogur, E.; YÜCEL, MUSTAFA; Öktem, Hüseyin Avni (Informa UK Limited, 2009-01-01)
To help in the development of Sunn Pest-resistant transgenic plants employing protease or alpha-amylase inhibitors, midgut hydrolytic enzymes of Sunn Pest (Eurygaster integriceps, Put.) (Heteroptera: Scutelleridae) were identified and characterized biochemically. We observed levels of very low proteolytic activity of trypsin (3 nmoles/min/mg), elastase (0.66 nmoles/min/mg) and leucine aminopeptidase-like (14.4 nmoles/min/mg) proteases, but no chymotrypsin and papain-like activity. Proteolytic activities wer...
