Comparative typing of L. delbrueckii subsp bulgaricus strains using multilocus sequence typing and RAPD-PCR

2011-09-01
Comparative typing analysis of 25 Lactobacillus delbrueckii subsp. bulgaricus strains, isolated from traditional yoghurts in Turkey, was performed by RAPD-PCR (randomly amplified polymorphic DNA-PCR) and MLST (multilocus sequence typing). RAPD-PCR analyses were performed using two primers; M13 and 1254. Primer 1254 produced better results than primer M13. The bands produced by primer 1254 were brighter and easier to interpret, and a higher number of bands were produced. In addition, clusters produced by primer 1254 were grouped according to the source of isolation. MLST analysis was performed using three genes, beta-gal, pheS and rpoA, and isolates were successfully characterized at strain level. To our knowledge, MLST analyses were used for the first time for strain level discrimination in L. delbrueckii subsp. bulgaricus. It enabled a detailed understanding of L. delbrueckii subsp. bulgaricus strains by using allele and sequence types' analysis. Both MLST and RAPD allowed for the typing of clusters according to the isolation source, while RAPD provided an increased differentiation. However, by increasing the number of genes analyzed, the discriminatory power of MLST could be increased.

Characterization of bacteriocins from two Lactococcus lactis subsp lactis isolates
Akcelik, O; Tukel, C; Özcengiz, Gülay; Akcelik, M (Wiley, 2006-03-01)
In this study, bacteriocins from two Lactococcus lactis subsp. lactis isolates from raw milk samples in Turkey designated OC1 and OC2, respectively, were characterized and identified. The activity spectra of the bacteriocins were determined by using different indicator bacteria including Listeria, Bacillus and Staphylococcus spp. Bacteriocins were tested for their sensitivity to different enzymes, heat treatments and pH values. Loss of bacteriocin activities after alpha-amylase treatment suggested that they...
Efficiency of pulse pressure treatment for inactivation of Escherichia coli and Listeria innocua in whole milk
BUZRUL, Sencer; Alpas, Hami; LARGETEAU, Alain; DEMAZEAU, Gerard (Springer Science and Business Media LLC, 2009-05-01)
Escherichia coli ATCC 11775 and Listeria innocua ATCC 33090 in whole milk were inactivated by single- and multi-pulsed (up to 10 pulses) high hydrostatic pressure (HHP) treatments. Both bacteria showed similar resistance to single- and multi-pulsed HHP. The efficiency of pulsed pressure treatment depends on the combination of holding time of each pulse and number of pulses. It was observed that multi-pulsed pressure treatment instead of traditional single-pulsed HHP could be used to pasteurize milk at a low...
Optimization of microwave baking of model layer cakes
Şümnü, Servet Gülüm; Bayindirli, L (Springer Science and Business Media LLC, 2000-01-01)
Response surface methodology was used to optimize the formulation of microwave-baked cakes. The independent variables were water content, emulsifier content baking time, oven power, shortening content and starch type. The quality factors evaluated were specific gravity of batter and volume index, uniformity index and tenderness of the crumb. Constraints for quality factors were obtained by conventional baking of American Association of Cereal Chemists high-ratio cake formulation. Multiple contour plots show...
Response surfaces of hazelnut oil yield in supercritical carbon dioxide
Ozkal, SG; Yener, Meryem Esra; Salgin, U; Mehmetoglu, U (Springer Science and Business Media LLC, 2005-01-01)
Response Surface Methodology was used to determine the effects of solvent flow rate (1, 3 and 5 g/ min), pressure (300, 375 and 450 bar) and temperature (40, 50 and 60 degreesC) on hazelnut oil yield in supercritical carbon dioxide (SC-CO2). Oil yield was represented by a second order response surface equation (R-2 = 0.997) using Box-Bhenken design of experiments. Oil yield increased with increasing SC-CO2 flow rate, pressure and temperature. The maximum oil yield was predicted from the response surface equ...
Optimization of halogen lamp-microwave combination baking of cakes: a response surface methodology study
Sevimli, KM; Şümnü, Servet Gülüm; Şahin, Serpil (Springer Science and Business Media LLC, 2005-07-01)
The main objective of this study was to optimize processing conditions during halogen lamp-microwave combination baking of cake by using response surface methodology. The independent variables were the power of the upper halogen lamp (50, 60, and 70%), the power of the lower halogen lamp (50, 60, and 70%), the power of the microwaves (30, 40, and 50%), and the baking time (4, 5, and 6 min). Weight loss, specific volume, color, and the texture profile of the cakes were determined. The upper halogen lamp powe...
