Projemiz kapsamında nükleik asit (DNA), protein ve hücre olmak üzere farklı moleküllerin tayininde kullanılabilecek dizi (array) platformlarının geliştirilmesi amaçlanmıştır. Geliştirilmesi hedeflenen platformların sandiviç hibridizasyon esasına dayalı olması ve farklı görüntüleme yöntemleri ile çalıştırılması hedeflenmiştir. Sandiviç hibridizasyon esasına dayalı DNA dizi platformunun geliştirilmesi esnasında cam yüzeylerin kaplanması, probların yüzeye tutturulması, adaptör DNA‟nın bağlanma koşullarının optimizasyonu, görüntüleme probunun hibridizasyon ve yıkama koşularının optimizasyonu ve florasan, quantum noktacık ve altın nanoparçaçık olmak üzere üç farklı görüntüleme yönteminin geliştirilmesi konusunda araştırmalar yürütülmüştür. Gerçekleştirilen deneyler sonucunda florasan prob ve kuantum noktacıkları ile görüntülenebilen, düşük arka plan sinyal seviyesine sahip ve oldukça verilmi çalışan bir dizi platformuna ulaşılmıştır. Projemizin diğer bölümlerinde aptamer tabanlı dizi platformlarında protein ve hücrelerin algılanabilmesi ile ilgili araştırmalar gerçekleştirilmiştir. Bu çalışmalarda da aptamerlerin yüzeye bağlanması, protein ve hücrelerin bağlanma koşullarının optimizasyonu ve florasan ve/veya kuantum noktacıkları kullanılarak görüntü alma konusunda araştırmalar yürütülmüştür. Bu çalışmalar sonucunda yüzeyde oluşturulan aptamer dizilere trombin proteinin özgün olarak bağlandığı ve bağlanan proteinlerin kuantum noktacıkları ile görüntülenebildiği gösterilmiştir. Hücre çalışmalarında da oluşturulan aptamer dizilerine lösemi kanser hücre hatlarının seçici olarak bağlandığı ve florasan işaretli ikincil aptamerler ile işaretlenebildiği gösterilmiştir. Ulaşılan bu sonuçlar protein ve hücre tanısında kullanılabilecek aptamer dizilerinin geliştirilebilmesi açısından konsept doğrulama niteliği taşımaktadır. Projemiz kapsamında iki yüksek lisans tez çalışması tamamlanmış, iki doktora tezinin de bazı bölümleri gerekleştirilmiştir. Proje faaliyetleri sonucunda grubumuz bünyesinde birçok yeni Ar-Ge projesi tetiklenmiş ve bunlara bağlı muhtelif tez çalışmaları başlatılmıştır.
