Staphylococcus aureus insan vücudu üzerinde sıklıkla asemptomatik olarak taşınan fırsatçı bir patojendir. Metisiline dirençli S. aureus (MRSA) sonradan edindikleri gen sayesinde metisilin ve genel olarak diğer tüm β-laktam antibiyotiklere karşı dirençlidir. MRSA dünyadayaygın bir patojen olup, diğer S. aureus enfeksiyonları ile karşılaştırıldığında MRSA enfeksiyonlarının daha yüksek oranda morbidite ve mortalite gösterdiği görülmektedir. Antibiyotikler ve diğer ilaçların doğal sularda ve yüzey sularında bulunması, nehirler gibi klinik olmayan ortamlarda yüksek direnç gösteren bakterilerin oluşmasına neden olmaktadır. Bunların içinden insanlar ve hayvanlar tarafından taşınan metisiline dirençli stafilokoklar ve β-laktam antibiyotiklere direnç sağlayan mecA geni taşıyan bakteriler özellikle dikkat çekmektedir. Metisiline dirençli stafilokokların yayılımlarındaki sürekli artışa rağmen, mecA genini taşıyan stafilokok ve diğer potansiyel patojen bakterileri rezervuar olarak barındıran doğal çevre hakkında çok az bilgi bulunmaktadır. Metisilin dirençli S. aureus penisilin bağlayıcı protein PBP2a içermesi ile oluşan hedef bölgelerin değiştirilmesi nedeniyle βlaktamlara karşı olan afinitesi azalmaktadır. Bu proteini kodlayan mecA geni mobil SCCmec olarak adlandırılan kromozom kaseti üzerinde yer almaktadır. Literatürde daha önce tarif edilmiş olan tüm SCCmec kasetindeki mec kompleks genleri (mecA, mecI, ve mecR1) genelde aynıdır. Bu genler β-laktam antibiyotiklere direnci sağlamaktadır. Ancak, tüm mecA klonları metisiline dirençli değildir ve MRSA populasyonundaki tüm direnç seviyeleri, çeşitli kromozomal faktörler tarafından module edilen PBP2a üretimindeki verimliliğe bağlıdır. Bu durum, MRSA direnç seviyelerinin fenotipik duyarlı ile yüksek dirençli arasında değişkenlik göstermesinin nedenini açıklamaktadır. Bu kaygılara dayanarak, bu projede şehirsel ve fekal kirlilikten etkilenen yüzey sularında mecA taşıyan bakterilerin izole edilmesi, tanımlanması, karakterizasyonu ve mecA spesifik DNA probları kullanarak mevsimsel olarak takip edilmesi planlanmıştır. Daha önce TÜBİTAK tarafından desteklenen 111T045 nolu proje kapsamında Kırıkkale il sınırları içerisinden geçen Kızılırmak’ın 12 farklı bölgesinden alınmış olan su örneklerinden metisilin dirençli bakteriler izole edilmiştir. Bu proje kapsamında ise, 111T045 nolu proje kapsamında izole edilen ve daha önceden stok kültürleri hazırlanmış olan bu bakterilerin tanımlanması, karakterize edilmesi, mecA genlerinin varlıklarının gösterilmesi planlanmış ve mecA spesifik DNA probları hazırlanarak 2011-2012 yılları süresince üçer aylık periyotlarla alınmış ve fikse edilmiş su örneklerinde mecA içeren bakterilerin mevsimsel olarak takip edilmesi planlanmıştır. Metisilin dirençli suşların tanımlanması 16S rRNA dizi iii analizi ve yağ asidi analizi ile yapılmıştır. Fikse edilmiş su örneklerindeki mecA geni taşıyan suşların populasyonlarındaki mevsimsel değişiklikler ise mecA hedefli floresan işaretli oligonükleotid problar kullanılarak floresan in situ hibridizasyon yöntemi ve görüntü analizi mikroskopu ile takip edilmiştir. Böylece mecA geninin S. aureus dışında başka bakterilerde de bulunup bulunmadığı ve mecA spesifik prob ile metisilin dirençli bakterilerin yüzey sularında takibinin yapılmasının uygunluğu belirlenmiştir. mecA hedefli floresan işaretli oligonükleotid problar kullanılarak floresan in situ hibridizasyon yöntemi ile metisilin dirençli bakterilerin yüzey sularında takip edilmesine yönelik literatürde herhangi bir çalışma bulunmamaktadır. Bu bakımdan projenin benzer konudaki çalışmalara da önemli katkıda bulunacağı düşünülmektedir.
