Evaluating Oxygen Tensions Related to Bone Marrow and Matrix for MSC Differentiation in 2D and 3D Biomimetic Lamellar Scaffolds
Sayin, Esen; Baran, Erkan Turker; Elsheikh, Ahmed; Mudera, Vivek; Cheema, Umber; Hasırcı, Vasıf Nejat (2021-04-01)
The physiological O-2 microenvironment of mesenchymal stem cells (MSCs) and osteoblasts and the dimensionality of a substrate are known to be important in regulating cell phenotype and function. By providing the physiologically normoxic environments of bone marrow (5%) and matrix (12%), we assessed their potential to maintain stemness, induce osteogenic differentiation, and enhance the material properties in the micropatterned collagen/silk fibroin scaffolds that were produced in 2D or 3D. Expression of ost...
Evaluation of metabolite variation between acute lymphoblastic leukemia (ALL) and chronic myeloidleukemia (CML) cell lines via triple quadrupole LC-MS
Pekşen, Ceren; Özen, Can.; Department of Biotechnology (2019)
Hematologic cancers have two origins: myeloid and lymphoid. While lymphatic leukemia, originate from the lymphoid cell line, acute myeloid leukemia (AML) and chronic myeloid leukemia (CML) originate from the myeloid cell line. CML is granulocyte cancer and in this disease, as well as granulocytes, the number of cells that cause blood clotting, which are called platelets, may increase in the blood. In CML, a non-hereditary genetic abnormality, Philadelphia chromosome, where a structural change occurs in chro...
Evaluation of Staphylococcus aureus DNA aptamer by enzyme-linked aptamer assay and isothermal titration calorimetry
BAYRAÇ, CEREN; Öktem, Hüseyin Avni (2017-02-01)
To monitor the specificity of Staphylococcus aureus aptamer (SA-31) against its target cell, we used enzyme-linked aptamer assay. In the presence of target cell, horseradish peroxidase-conjugated streptavidin bound to biotin-labeled SA-31 showed specific binding to S aureus among 3 different bacteria with limit of detection of 103 colony-forming unit per milliliter. The apparent K-a was 1.39 mu M-1 +/- 0.3 mu M-1. The binding of SA-31 to membrane proteins extracted from cell surface was characterized using ...
ASSESSING PERLITE AS A SAND SUBSTITUTE IN FILTRATION
ULUATAM, SS (1991-06-01)
Quartz sand is commonly used in filter beds to remove suspended solids. In Turkey, however, sources of sand suitable for filtration are limited and costly, making it necessary for utilities to examine alternative bed materials. Crushed raw perlite, which is plentiful along Turkey's Aegean Coast, was examined for its suitability as a substitute for sand. Sieve analysis, porosity, particle density, and solubility in 20 percent concentrated HCl acid test results as well as the costs of perlite and sand were...
