Electrophoretic analysis of genetic variability in the house fly ( Musca domestica L. )

Farklı karasinek (Muşça domestica L.) soylarındaki genetik varyasyonun miktarı ve soylar arasındaki genetik farklılaşmanın derecesi iki enzim sisteminin (maleyt dehidrogenaz (E.C 1.1.1.37, Mdh) ve hekzokinaz (E.C 2.7.1.1, Hk)j ızoenzimlerinin vertikal poliakrıla- r mit jel elektroforezi tekniği ile çalışılması sonucunda ölçüldü. Üç farktı karasinek soyunda Mdh için iki farklı aktivite bölgesi, Hk için ise dört farklı aktivite bölgesi gözlendi. Her iki enzim sistemi için kalıtım analizleri yapılarak her bölgenin bir lokus ve iki allel tarafından kontrol edildiği gösterildi. Soylar içindeki genetik varyas-yonun ve soylar arasındaki genetik farklılaşmanın derecesini kantitatif olarak gösteren parametreler herbır lokus için hesaplanan allel frekans-ları kullanılarak elde edildi. Bazı lokuslarda gözlenen heterozigotlu-ğun beklenenden önemli derecede az olması akrabalar arası çiftleşmenin ortaya çıkardığı bir durum olarak yorumlandı. Soyların beklenen ortala-ma neterozigotl uk değerleri (0.400 - 0.475) benzer bulundu. Birbiri ile çok yakın iki soy arasındaki genetik uzaklık 0.0025. olarak hesaplandı.Çalışılan soylar arasındaki genetik uzaklıklar (0.0025 - 0.0401) genellikle lokal populasyonlar arasında bulunan genetik uzaklıkların sınırları içinde.
