Loss of the Nuclear Envelope Protein LAP1B Disrupts the Myogenic Differentiation of Patient-Derived Fibroblasts
Kayman Kürekçi, Gülsüm; Acar, Aybar Can; Dinçer, Pervin R. (2022-11-01)
Lamina-associated polypeptide 1 (LAP1) is a ubiquitously expressed inner nuclear membrane protein encoded by TOR1AIP1, and presents as two isoforms in humans, LAP1B and LAP1C. While loss of both isoforms results in a multisystemic progeroid-like syndrome, specific loss of LAP1B causes muscular dystrophy and cardiomyopathy, suggesting that LAP1B has a critical role in striated muscle. To gain more insight into the molecular pathophysiology underlying muscular dystrophy caused by LAP1B, we established a patie...
Regulation and role of CCAAT/enhancer binding protein during differentiation of intestinal epithelial cells
Memişoğlu, Aslı Sade; Banerjee, Sreeparna (2013-09-01)
Background: The molecular mechanisms of differentiation of intestinal epithelial cells, is poorly understood and disruption of this balance may result in neoplastic transformation and malignant growth. The family of CCAAT/Enhancer Binding Protein (C/EBP) transcription factors is implicated in cellular growth, differentiation, inflammation and development and control differentiation in several cell types. The involvement of these transcription factors in intestinal differentiation is not known. The aim of th...

Oligomerization and cell surface expression of recombinant GABA(A) receptors tagged in the delta subunit
Oflaz, Furkan Enes; Son, Çağdaş Devrim; Arslan, Ayla (IMR Press, 2019-12-01)
The gamma-Aminobutyric acid type A receptors (GABA(A) Rs) are heteropentameric chloride channels responsible for primary inhibition in the mammalian brain. Studies have shown the expression of recombinant GABA(A) R subunits tagged with the green fluorescent protein (GFP), a 26.9 kDa protein that exhibits bright green fluorescence when exposed to light in the blue to ultraviolet range. This allows the formation of recombinant proteins essential for the development of relevant in-vitro and in-vivo methodologi...
Induction of apoptosis and cell cycle arrest on U266 multiple myeloma cell line by prochlorperazine
Hüsnügil, Hepşen Hazal; Özen, Can; Banerjee, Sreeparna; Department of Biochemistry (2016)
Multiple myeloma (MM) is a plasma cell neoplasm accounting for 1% of all malignancies and 13% of hematological malignancies. Despite the introduction of potent anticancer agents, MM remains as an incurable disease. High frequency of relapses and acquisition of resistance to current chemotherapy create a need for the development of novel agents for MM treatment. Prochlorperazine (PCP) is an FDA-approved phenothiazine drug, mainly used for the treatment of chemotherapy-associated nausea and vomiting. In addit...
