Deterioration of retina and death of the retinal cells due to age, diabetes, or occlusion can cause retinal degeneration which leads to loss of vision. In this study, it is aimed to design a bilayered matrix to mimic the choroid and the Bruch's membrane of the retinal tissue. As choroid, a microchanneled network resembling a fractal tree design was fabricated by photolithography over photo-cross-linkable methacrylated hyaluronic acid hydrogel. Gelatin or collagen was immobilized into the microchannels to enhance adherence of Human Umbilical Vein Endothelial Cells (HUVEC). At late culture periods (2 weeks), formation of tubular structures due to proliferation of the attached cells was observed. As Bruch's membrane, an electrospun fibroin nanofiber mat was produced to grow retinal pigment epithelium (RPE) cells on. Cellular interactions between RPE and HUVEC in the microchannels were investigated in a coculture model in a noncontact mode. It was deduced that by combining the RPE layer on the highly permeable Bruch's membrane with the choroid layer populated by HUVECs, a retinal substitute which might have a potential for use in the treatment of retinal diseases is possible. Characterization of Novel O-Glycans Isolated from Tear and Saliva of Ocular Rosacea Patients
Özcan Kabasakal, Süreyya; Vieira, Ana C.; Park, Gun Wook; Kim, Jae Han; Mannis, Mark J.; Lebrilla, Carlito B. (American Chemical Society (ACS), 2013-03-01)
O-Glycans in saliva and tear isolated from patients suffering from ocular rosacea, a form of inflammatory ocular surface disease, were profiled, and their structures were elucidated using high resolution mass spectrometry. We have previously shown that certain structures, particularly sulfated oligosaccharides, increased in the tear and saliva of rosacea patients In this study, the structures of these glycans were elucidated using primarily tandem mass spectrometry. There were important similarities in the ...
Construction of retina substitute by using tissue engineering approach /
Kömez, Aylin; Hasırcı, Vasıf Nejat; Baran, Erkan Türker; Department of Biotechnology (2015)
Millions of people are affected by retinal diseases such as age-related macular degeneration (AMD) and retinitis pigmentosa (RP, night blindness). Retinal tissue engineering has potential to become a valid therapeutic approach to regenerate retina. The aim of this study was to construct a biomimetic retinal tissue substitute by using biocompatible and natural polymers and in vitro techniques. For this purpose, a retinal pigment epithelial (RPE) cell monolayer was formed on an electrospun silk fibroin mesh t...
Influence of structural length-scale variations on azimuth-resolved light scattering patterns of inhomogeneous cell models
Arifler, Dizem; Guillaud, Martial (2015-06-24)
Optical scattering provides an intrinsic contrast mechanism for the diagnosis of early precancerous changes in tissues. There have been a multitude of numerical studies targeted at delineating the relationship between cancer-related alterations in morphology and internal structure of cells and the resulting changes in their optical scattering properties. Despite these efforts, we still need to further our understanding of inherent scattering signatures that can be linked to precancer progression. As such, c...
Improving the sub-cortical gm segmentation using evolutionary hierarchical region merging
Çiftçioğlu, Mustafa Ulaş; Gökçay, Didem; Department of Medical Informatics (2011)
Segmentation of sub-cortical Gray Matter (GM) structures in magnetic resonance brain images is crucial in clinic and research for many purposes such as early diagnosis of neurological diseases, guidance of surgical operations and longitudinal volumetric studies. Unfortunately, the algorithms that segment the brain into 3 tissues usually suffer from poor performance in the sub-cortical region. In order to increase the detection of sub-cortical GM structures, an evolutionary hierarchical region merging approa...
Review of MRI-based Brain Tumor Image Segmentation Using Deep Learning Methods
Işın, Ali; Direkoğlu, Cem; Şah, Melike (Elsevier BV; 2016)
Brain tumor segmentation is an important task in medical image processing. Early diagnosis of brain tumors plays an important role in improving treatment possibilities and increases the survival rate of the patients. Manual segmentation of the brain tumors for cancer diagnosis, from large amount of MRI images generated in clinical routine, is a difficult and time consuming task. There is a need for automatic brain tumor image segmentation. The purpose of this paper is to provide a review of MRI-based brain ...
