High hydrostatic pressure (HHP) as a non-thermal technology is an effective tool for microbiologically safe and shelf-stable fruits Mycotoxin citrinin (CIT) is a toxic secondary metabolite, especially produced from filamentous fungus Penicillium citrinum and is also produced by other species of Penicillium. Aspergillus, and Monascus that are able to develop on olive after harvest, during brine and storage of olives Nutritional benefits of olive fruit are mainly related to phenolics such as hydroxytyrosol, oleuropein and antioxidative effects. With HHP application of olives, total mold was reduced to 90% at 25 degrees C whereas it was 100% at 4 degrees C based on Rose-Bengal Chloramphenicol Agar (RBCA) Total Aerobic-Mesofilic Bacteria load was reduced to 35-76% at 35 +/- 2 degrees C based on the Plate Count Agar (PCA). Citrinin load was reduced to 64-100% at 35 +/- 2 degrees C 2.5, 10, 25, and 100 ppb of spiked citrinin in sample were degraded as %56; %37, %9, and %1 3, respectively. 2 5 ppb and less citrinin contamination in table olive were degraded more (56%). Total phenolics were increased to 2 1-2 5-fold after HHP (as mgGA/100 g) Hydroxytyrosol in olives increased on average 0 8-2.0-fold whereas oleuropein decreased on average 1-1.2-fold after HHP (as mg/kg dwt) Antioxidant activity values varied from 17 238 to 29.344 mmol Fe(2+)/100 g for control samples whereas 18.579-32 998 mmol Fe(2+)/100 g for HHP-treated samples HHP could be used in the olive industry as non-thermal preservation (C) 2009 Elsevier Ltd All rights reserved.

Antioxidant and cytotoxic properties of Salvia Absconditiflora and effects on CYP1A1, CYP1B1 gene expressions in breast cancer cell lines
Yılmaz, Selis; İşcan, Mesude; Güray, Tülin; Department of Biochemistry (2013)
Salvia genus is a widely cultivated genus and used in medicine for various purposes as having antimicrobial, antioxidant, anticarcinogen and anti-inflammatory features. In this study the aim was to investigate phenolic composition of Salvia absconditiflora and understand the possible effects of those constituents in cancer related drug metabolizing enzymes. Salvia absconditiflora showed 80,43 % Radical Scavenging Activity against DPPH radical. Total flavonoid content was found as one third of total phenolic...
In vitro activity of panomycocin, a novel exo-beta-1,3-glucanase isolated from Pichia anomala NCYC 434, against dermatophytes
Izgu, Fatih; Altinbay, Demet; Tureli, Akif Emre (2007-01-01)
Killer proteins that are produced and secreted into the environment by certain yeast strains are considered as promising antifungal agents. In this study, in vitro activity of Pichia anomala NCYC 434 (K5) killer protein, panomycocin, which is a 49 kDa glycoprotein with an exo-beta-1,3-glucanase activity was tested against 41 isolates of dermatophytes. Minimum inhibitory concentrations (MICs) were determined by a broth microdilution method based on the reference document M38-A of Clinical and Laboratory Stan...
Overexpression of serine alkaline protease encoding gene in Bacillus species: performance analyses
Çalık, Pınar; Ozdamar, TH (Elsevier BV, 2003-12-02)
Bacillus species carrying subC gene encoding serine alkaline protease (SAP) enzyme were developed in order to increase the yield and selectivity in the bioprocess for SAP production. For this aim, subC gene was cloned into pHV1431 Escherichia coli-Bacillus shuttle vector, and transferred into nine host Bacillus species, i.e. B. alvei, B. amyloliquefaciens, B. badius, B. cereus, B. coagulans, B. firmus, B. licheniformis, B. sphaericus and B. subtilis. The influence of the host Bacillus species on SAP product...
Utilization of pretreated molasses for serine alkaline protease production with recombinant Bacillus species
Calik, G; Pehlivan, N; Kalender, N; Ozdamar, TH; Çalık, Pınar (Informa UK Limited, 2003-05-01)
Recombinant Bacillus amyloliquefaciens, Bacillus cereus, Bacillus subtilis , and Bacillus licheniformis were used for the production of serine alkaline protease (SAP) utilizing chemically and/or physically pretreated molasses. The highest enzyme activity was obtained with r- Bacillus subtilis , with the complex medium involving physically treated molasses having 20 kg m(-3) initial sucrose concentration in small-scale, agitation- and heating rate-controlled bioreactors at t=63 h. Effects of oxygen transfer ...
Microbial detoxification of groundnut meal naturally contaminated with aflatoxin using rhodococcus erythropolis
Doğan, Önay Burak; Çekmecelioğlu, Deniz; Bozoğlu, Faruk; Department of Food Engineering (2015)
Aflatoxins are highly mutagenic toxins with carcinogenic effects produced as secondary metabolites by fungal species Aspergillus flavus and Aspergillus parasiticus under certain conditions. Chronic or acute consumption of aflatoxins found in food and feed products possesses great health risks. It is particularly an important problem in animal feed from food waste and by-products. Therefore there is growing need to eliminate aflatoxins from contaminated products. In this study, first the optimum growth condi...
