beta-mannanases can degrade galactomannans to mannose and it has been used in various application areas. The aim of this study was to produce the beta-mannanase from carob pod extract including different nitrogen sources. The best operation values for fermentation were determined to be 8% initial sugar concentration with 0.5% yeast extract, 100 r.p.m., and 7% inoculation rate, which yielded the maximum beta-mannanase activity as 423.60 U ml(-1). Effects of nitrogen sources on beta-mannanase activity were also studied and it reached 695.6 U ml(-1) by using 0.5% of ammonium nitrate as the nitrogen source at the determined optimum conditions. Results also showed that meat bone meal and soybean meal could be used as cost effective nitrogen sources based on achieved beta-mannanase activity.

Characterization of local flow behaviour in conical spouted beds
Yeniçeri, Ali; Külah, Görkem; Department of Chemical Engineering (2019)
Conical spouted beds are used in various physical and chemical applications such as drying, coating, and pyrolysis of waste plastic and biomass owing to their effective feature of gas-solid contact. To design and operate these units in high efficiency, a complete understanding of the phenomena and detailed investigation of local flow structure are of paramount importance. In this study, the effects of bed design parameters (unit size, conical angle, spouting gas-inlet diameter) and operational conditions (p...
The effect of Dilution, Aeration, and Agitation on Fungal Cellulase and Xylanase Production by DDGS-based Fermentation Media in Stirred Tank Bioreactors
Iram, Attia; Çekmecelioğlu, Deniz; Demirci, Ali (2022-01-01)
Fungal cellulases and xylanases are commonly used for biological pretreatment of lignocellulosic biomass into simple sugars, which can then be converted into many value-added products including bioethanol. However, these enzymes are costly to produce and need a feedstock that should be more economical and high in fiber and other nutrients. Distillers dried grains with solubles (DDGS) is the byproduct of corn ethanol production and can be considered an appropriate feedstock. Among various problems in adaptat...
Attenuation of polychlorinated biphenyls (PCBs) under anaerobic conditions
Kaya, Devrim; Sanin, Faika Dilek; İmamoğlu, İpek; Department of Environmental Engineering (2012)
Polychlorinated biphenyls (PCBs) are toxic and persistent anthropogenic contaminants. Concern on their adverse health effects has led to their regulation in air, water and/or soil in addition to sludge. Hence, removal of PCBs in various matrices, including transformer oils (TO) is a priority. This study aims to investigate PCB-118 and Aroclor 1254 toxicity and dechlorination by varying certain critical experimental components including electron donor (sludge or fatty acids), inocula (unacclimated or acclima...
Portable Bioactive Paper-Based Sensor for Quantification of Pesticides
Kavruk, Murat; Ozalp, Veli Cengiz; Öktem, Hüseyin Avni (Hindawi Limited, 2013-01-01)
A paper-based biosensor was developed for the detection of the degradation products of organophosphorus pesticides. The biosensor quantifies acetylcholine esterase inhibitors in a fast, disposable, cheap, and accurate format. We specifically focused on the use of sugar or protein stabilizer to achieve a biosensor with long shelf-life. The new biosensor detected malathion with a detection limit of 2.5 ppm in 5 min incubation time. The operational stability was confirmed by testing 60 days storage at 4 degree...
LIFE CYCLE ASSESSMENT OF ACTIVATED CARBON ADSORPTION AND OZONATION TO REMOVE MICROPOLLUTANTS FROM TEXTILE WASTEWATER
Akdeniz, Ayşe Sena; Yetiş, Ülkü; Department of Environmental Engineering (2022-8-29)
To avoid potential adverse effects by micropollutants in secondary textile wastewater, it is necessary to apply an advanced treatment process, among which activated carbon adsorption and ozone oxidation are the most attractive alternatives. There is still a lack of studies evaluating the environmental impacts of these processes for the tertiary treatment of textile wastewater. Using the life cycle assessment (LCA) methodology, this study assesses and compares the environmental impacts of ozone oxidation and...
