All rights reserved. Background and purpose: Synthetic fertilizers damage the environment. Biofertilizers that consist of microorganisms emerge as an environmentally friendly alternative. Biofertilizers improve plant growth by mobilizing soil nutrients, triggering plant hormone synthesis, or competing with pathogenic bacteria. However, biofertilizers often fail due to insufficient colonization of the plant roots. Materials and methods: To explore the colonization dynamics of a bacterial strain commonly used in biofertilizers, Bacillus subtilis OSU-142 (OSU-142), developing a set of primers specific to OSU-142 was aimed. Since its genome is unknown, to identify genomic regions unique to OSU-142 strain, DNAs of more than 40 bacterial strains were fingerprinted, most of which belong to Bacillus subtilis using the Randomly Amplified Polymorphic DNA (RAPD) method. Results: This approach identified a polymorphic band at 880 bp, which was then cloned and sequenced. The sequence showed no perfect match to any known sequences in the tested genomic databases indicating that the region of OSU-142 DNA is highly unique. The primer was converted to Sequence-Characterized Amplified Region (SCAR) primers, and its functionality in detecting OSU-142 genome was confirmed. Newly designed primer set can specifically detect OSU-142. Conclusions: These primers can be useful for basic science or commercial applications on tracking OSU-142 in various environments, thus contribut-ing to biofertilizers’ adoption in the long run.

Production of hemicellulose-based biodegradable films from agricultural wastes
Bahcegul, E.; Özkan, Necati; Bakir, U. (2009-09-01)
Owing to the increasing environmental awareness and restrictions together with the depletion of fossil-based resources, production of biodegradable films receives increasing attention to substitute at least a part of the largely consumed conventional synthetic plastics. Despite there are a variety of polymers that are used for biodegradable plastic production these alternatives are either more expensive compared to their non-biodegradable petroleum-based counterparts or obtained from sources such as corn or...
Development of fluorescent whole-cell bacterial bioreporters for detection of inorganic arsenic and cadmium
Elçin, Evrim; Öktem, Hüseyin Avni; Department of Biotechnology (2019)
Environmental heavy metal contamination in many regions of the world is a serious problem of ecological health. Fast and constant monitoring of their levels is significant for both preventing their accumulation and taking an immediate action for removal. As an alternative approach to standard laboratory techniques, various biosensor systems for detection of heavy metals have been proposed. Bacterial biosensors hold great promise for in-field detection of heavy metals. In this study, two different whole cell...
Evaluation of Organochlorine Pesticides (OCPs) and Polychlorinated Biphenyls (PCBs) Concentrations in the River and Marine Sediments of Samsun Coastline

Today, the persistent organic pollutant loads which are formed as a result of modern agriculture and industrial production are becoming very dangerous for the environment and human health. Exposure of living things to these pollutants can occur by direct exposure to pollutants emission or by transport of pollutants as a result of food chain and meteorological events. For this reason, all environmental samples should be analyzed in this context and risk assessments should be made especially for regions where...
Investigation of adsorption of pesticides by organo-zeolite from wastewater
Lüle, Güzide Meltem; Atalay, M Ümit; Özbayoğlu, Gülhan; Department of Mining Engineering (2011)
The aim of this study was to determine the adsorption capacity of activated carbon and organo-zeolites for removal of pesticides in water. In order to prepare organo-zeolite, two kinds of cationic surfactants, namely, hexadecyltrimethyl ammonium bromide (HTAB) and dodecyltrimethyl ammonium bromide (DTAB) were used. Adsorption studies of cationic surfactant on zeolite were investigated in respect to initial concentration of cationic surfactant, time, and temperature. It has been found that the best fitted is...
Investigation of alkaline pretreatment parameters on amulti-product basis for the co-production of glucose and hemicellulose based films from corn cobs
Toraman, Hilal Ezgi; Bakır Bölükbaşı, Ufuk; Özkan, Necati; Department of Chemical Engineering (2012)
There is an increasing trend in the world for using renewable sources of fuels and chemicals due to the continuous depletion of fossil fuel reserves besides the environmental issues related with the exploitation of these resources. Lignocellulosic biomass is seen as the most promising candidate to be used instead of fossil sources because of its availability, relatively low price and less competition with food and feed crops. In this study, corn cobs, a lignocellulosic agricultural waste, were subjected to ...
