Determination of doxorubicin amount conjugated to mPEG-b-PCL copolymer via pH sensitive hydrazone bond pH duyarlı hidrazon bağıyla mPEG-b-PCL kopolimerine konjuge edilen doksorubisin miktarının belirlenmesi

Turk Hijyen ve Deneysel Biyoloji Dergisi. All Rights Reserved. Objective: The aim of this study was to find the efficient medium to bind an anticancer drug, Doxorubicin (DOX), to a synthesized polymer, methoxy poly(ethylene glycol)-block-polycaprolactone (mPEG-b-PCL) via pH sensitive hydrazone bonds and to determine the amount of conjugated drug. Methods: DOX conjugation was carried out in two different media [dimethyl sulfoxide (DMSO) and methanol-trifluoroacetic acid (MeOH-TFA)]. The amount of conjugated drug was determined with two different methods. One method was applied the dissolution of the conjugate in chloroform: methanol (Ch: MeOH, 1: 1 v/v) solution without considering pH responsiveness, and the other method was after breaking pH sensitive hydrazone bonds in acidic medium [using three different media as 0.1 M hydrochloric acid (HCl), concentrated HCl (12 M HCl) and concentrated sulfuric acid (18.3 M H2SO4)]. Results: The highest conjugation efficiency was obtained when the conjugation was achieved in MeOHTFA solution, and for the polymer-drug conjugates after the treatment with concentrated sulfuric acid. Conclusion: It was concluded that, MeOH-TFA method was a good method for conjugation of DOX to mPEG-b-PCL copolymer, and H2SO4 (18.3 M) method was better than any other present in literature for determination of the amount of DOX linked to the polymer via hydrazone conjugation.

Synthesis and characterization of fluorine-substituted polyanilines
Cihaner, A; Önal, Ahmet Muhtar (Elsevier BV, 2001-09-01)
Poly(2-fluoroaniline), P2FAN, poly(3-fluoroaniline). P3FAN. and poly(4-fluoroaniline), P4FAN, have been synthesized from fluorine substituted aniline monomers in aqueous acidic medium using potassium dichromate as oxidizing agent. Characterization of polymer products has been carried out using FTIR, and NMR spectroscopic techniques. Thermal analysis of polyfluoroaniline powders was also investigated using differential scanning calorimetry and thermogravimetric analysis. To compare the structural properties ...
Bio-functional analysis of RAD23 gene using virus induced gene silencing method in response to powdery mildew attack in Barley
Aksoy, Yağmur; Akkaya, Mahinur S.; Ersoy, Figen; Department of Biotechnology (2012)
Blumeria graminis f.sp. hordei is an obligate biotrophic parasite causing powdery mildew disease in several cereals, including barley. Virus induced gene silencing (VIGS) is a widely used powerful tool in plant functional-genomic studies to determine the functions of genes in disease resistance. In this thesis study, the function of RAD23 gene in barley in plant resistance was investigated. The RAD23 is a ubiquitin (Ub) binding protein involved in programmed protein degradation by transferring of ubiquityla...
In situ monitoring of vancomycin resistance genes in surface waters by oligonucleotide probes
Nakipoğlu, Mustafa; İçgen, Bülent (null; 2015-09-13)
Vancomycin and teicoplanin resistance is a major cause of nosocomial infections worldwide. It is caused by various van genes located on either chromosome or transposons such as Tn1549 and its homologues. They encode proteins responsible of the moiety change on terminal pentapeptide bridge in peptidoglycan layer that eventually hinders the effectiveness of vancomycin up-to 1000 times. Although van genes are mostly found on enterococci, their homologues with high sim...

We present timing analysis of accreting X-ray pulsar GX 1+4 using the RXTE-PCA observations between 2001 and 2002. By constructing a phase connected timing solution for the source, we measured their spin frequencies and updated the long-term pulse frequency history of this source. We also reviewed the relation between frequency derivative and X-ray flux during this episode.
Identification of gene mutations ınvolved in drug resistance in liver cancer using rna-seq data analysis
Shojeai, Mona; Akhan, Ece; Acar, Aybar Can; Atalay, Rengül (null; 2017-06-28)
Cancer is the leading cause of death worldwide and the risk increases with aging. A significant concern in cancer research is the detection of cancer drug resistance associated somatic mutations. According to Global Cancer Statistics (GCS) liver cancer is the 5th most common and 2nd deadliest cancer in the world. Over the past two decades, the death rate for liver cancer increased by 2.5% per year and incidents were 3 times higher in men than in women (American Cancer ...
