Trientine is a chelator that specifically binds Cu(II) ions. Trientine hydrochloride salt is used in the treatment of Wilson’s disease, which is a genetic disorder of copper metabolism. This chelator molecule has significant potential to be used in other areas due to its high selectivity toward copper ions and low toxicity. In this thesis, trientine was synthesized using a peptide synthesis approach, a novel method for its preparation. Furthermore, 21 different trientine derivatives were synthesized through peptide synthesis to enhance its bioavailability, and subsequently reduced with a borane reagent. The dissociation constants of these 21 derivatives with copper ions were measured using fluorescence spectroscopy. The measured dissociation constants of the derivatives were nearly equivalent, and these values are consistent with theoretical calculations.
