Heterologous expression, characterization, and optimization of production of alpha-galactosidase from aspergillus fumigatus in aspergillus sojae
Gürkök, Sümeyra; Ögel, Zümrüt Begüm; Bölükbaşı, Ufuk; Department of Biotechnology (2012)
α-Galactosidase is an exo-glycosidase that hydrolyses non-reducing, α-1,6-linked α-galactose units from oligosaccharides, galactomannans, and galactolipids. α-Galactosidase activity has biotechnological, industrial, and medical importance. α-Galactosidase from A. fumigatus IMI 385708, in particular, can catalyse unique hydrolysis and transgalactosylation reactions on polymeric substrates. In this study, α-galactosidase of the human pathogen A. fumigatus IMI 385708 was first produced in a GRAS organism, Aspe...
Identification and characterization of hydrolytic enzymes of sunn pest (eurygaster integriceps) and cotton bollworm (helicoverpa armigera)
Özgür, Ebru; Öktem, Hüseyin Avni; Department of Biochemistry (2006)
In this study, hydrolytic enzymes from sunn pest (Eurygaster integriceps) and cotton bollworm (Helicoverpa armigera) midguts were identified and characterized in terms of their optimum pH, Km and Vmax values. Hydrolytic activities were also tested for inhibition by several protease and alpha-amylase inhibitors which can be used for the development of insect resistant plants through transgenic technologies. For sunn pest midgut, a low proteolytic activity, belonging mostly to trypsin-like and leucine aminope...
Removal of cobalt from zinc sulfate solution by cementation prior to zinc electrowinning
Kayın, Pınar Burcu; Topkaya, Yavuz Ali; Department of Metallurgical and Materials Engineering (2003)
The aim of this study was to investigate the removal of cobalt from zinc sulfate solution by cementation with the help of conventional and new type of additives that were 4% Sn-zinc alloy powder and 10% Sn-zinc alloy powder, respectively. Synthetic leach solutions containing 150 g/l Zn and 75 mg/l Co were prepared and used in all of the experiments. The parameters researched with the conventional method were the amount of arsenic trioxide and the effect of copper sulfate on cementation of cobalt. While usin...
Immobilization of glucoamylase onto activated pHEMA/EGDMA microspheres: properties and application to a packed-bed reactor
Arica, MY; Alaeddinoglu, NG; Hasırcı, Vasıf Nejat (1998-02-15)
Glucoamylase was covalently immobilized onto pHEMA/EGDMA microspheres of two different sizes: 50-100 mu m and 100-200 mu m in diameter. The activity of the enzyme on smaller microspheres was found to be almost twice that of the larger microspheres. A higher enzyme lending was observed on small microspheres (0.64 mg g(-1) support) as compared to large spheres (0.40 mg g(-1) support). The K-m of glucoamylase was significantly increased (approximately five times) upon immobilization, indicating decreased affin...
