Purification and characterization of an intracellular chymotrypsin-like serine protease from Thermoplasma volcanium
Kocabıyık, Semra (2006-01-01)
An intracellular serine protease produced by Thermoplasma (Tp.) voleanium was purified using a combination of ammonium sulfate fractionation, ion exchange, and et-casein agarose affinity chromatography. This enzyme exhibited the highest activity and stability at pH 7.0, and at 50 degrees C. The purifed enzyme hydrolyzed synthetic peptides preferentially at the carboxy terminus of phenylalanine or leucine and was almost completely inhibited by PMSF, TPCK, and chymostatin, similarly to a chymotrypsin-like ser...
Preparation and microstructure of sol-gel derived silver-doped silica
Akkopru, Betul; Durucan, Caner (2007-08-01)
Silver-doped silica was prepared by hydrolysis and condensation of tetraethyl orthosilicate (TEOS, Si(OC2H5)(4)) in the presence of a silver nitrate (AgNO3) solution by two different synthesis methods. In the first synthesis route, sol-gel mixtures were prepared using an acid catalyst. In the second synthesis route, silver-doped silica gels were formed by two-step acid/base catalysis. For the same concentration of silver dopant [AgNO3]/[TEOS] = 0.015 acid-catalyzed sol-gel formed a microporous silica with a...
PURIFICATION AND CHARACTERIZATION OF 2 FORMS OF SOLUBLE NADH CYTOCHROME-B5 REDUCTASES FROM HUMAN ERYTHROCYTES
ARINC, E; Güray, Nülüfer Tülün; SAPLAKOGLU, U; Adalı, Orhan (Elsevier BV, 1992-01-01)
1. Two forms of soluble NADH cytochrome b5 reductase were purified from human erythrocytes. Two distinct fractions both having the NADH cytochrome b5 reductase activity eluted from the second DEAE-cellulose column were further purified by ultrafiltration and 5'-ADP-agarose affinity chromatography.
Identification and characterization of hydrolytic enzymes of sunn pest (eurygaster integriceps) and cotton bollworm (helicoverpa armigera)
Özgür, Ebru; Öktem, Hüseyin Avni; Department of Biochemistry (2006)
In this study, hydrolytic enzymes from sunn pest (Eurygaster integriceps) and cotton bollworm (Helicoverpa armigera) midguts were identified and characterized in terms of their optimum pH, Km and Vmax values. Hydrolytic activities were also tested for inhibition by several protease and alpha-amylase inhibitors which can be used for the development of insect resistant plants through transgenic technologies. For sunn pest midgut, a low proteolytic activity, belonging mostly to trypsin-like and leucine aminope...
Characterization of the polymer of a dipyrrolyl monomer by pyrolysis mass spectrometry
Ertas, Merve; Hacaloğlu, Jale; Toppare, Levent Kamil (2004-08-01)
In this work, characterization of a homopolymer of succinic acid bis(4-pyrrol-1-ylphenyl) ester prepared by galvonastatic polymerization was carried out by direct pyrolysis mass spectrometry. Although decomposition of the monomer yielding mainly butadionic acid and pyrrole occurred under the galvonastatic polymerization conditions, growth of the polymer through the pyrrole moieties was also achieved, yielding a ladder-type polymer film. The polypyrrole chains contained both quinoid and aromatic units as in ...
