Embryogenic tissues (ET) of Turkish red pine (Pinus brutia TEN) were initiated from immature precotyledonary zygotic embryos sampled from 15 different plus trees. Seven collections were made weekly from June 10 to July 22, 2003. DCR basal medium supplemented with 13.6 mu M 2,4-dichlorophenoxyacetic acid (2,4-D) and 2.2 mu M benzylaminopurine (BAP) was used for initiation and maintenance of ET. Overall initiation frequency of ET in the study was 11.6%, initiation rates ranging between 4.7% and 24.1% per tree. Out of 12,940 explants tested, 3.4% were converted into established cell lines (ECL) following five subcultures. Of the maturation treatments tested, 80 mu M ABA, sucrose (3%) and maltose (3% and 6%), and 3.75% PEG combined with 1% gellan gum were the most suitable combination for somatic embryo maturation.

The effect of hybrid vigor on callus induction and plant regeneration from mature embryo culture of barley (Hordeum vulgare)
Ozgen, M; Birsin, MA; Onde, S (Springer Science and Business Media LLC, 2005-07-01)
The influence of hybrid vigor on callus induction, callus weight, regeneration capacity of callus, culture efficiency and number of regenerated plants from mature embryo cultures of winter barley ( Hordeum vulgare L.) hybrid was studied. A total of 15 F-1 hybrids and their parents were used for mature embryo culture. The statistical analysis of the results revealed that positive heterosis was noticed for all these characters except for callus weight. Plants regenerated in vitro were successfully established...
Microsatellite fingerprinting of homonymous grapevine (Vitis vinifera L.) varieties in neighboring regions of South-East Turkey

Genotyping of Turkish grapevine (Vitis vinifera L.) germplasm was characterized by use of six highly polymorphic microsatellite loci (VVS2, VVMD5, VVMD7, VVMD27, VrZAG62, VrZAG79). In this study we aimed to clarify the relationships between homonymous varieties coming from different regions. Our results showed a large degree of genetic variability among most of the homonymous cultivars. The number of alleles per locus ranged from 10 to 21, and gene diversity (expected heterozygosity) values ranged from 0.85...
Induction of embryogenic tissue from immature zygotic embryos in Pinus nigra J.F. Arnold subsp. nigra var. caramanica (Loudon) Businsky
Özkurt, Zeynep; Yildirim, Tolga; Önde, Sertaç; Kaya, Zeki (2008-06-12)
Embryogenic cultures (EC) of Anatolian black pine (Pinus nigra J.F.Arnold subsp. nigra var. caramanica (Loudon) Businsky) were initiated from immature pre-cotyledonary zygotic embryos sampled weekly from 16 different trees in 2 consecutive years. Douglas-fir cotyledon revised (DCR) medium supplemented with 13.6 μM of 2,4-D and 2.2 μM of BAP was used for initiation and maintenance of EC. Overall initiation frequencies of EC in the study were 0.92% in 2004 and 1.96% in 2005: tree values ranged from 0.0% to 7....
Genetic variability among Turkish pop, flint and dent corn (Zea mays L. spp. mays) races: Morphological and agronomic traits
Ilarslan, R; Kaya, Zeki; KANDEMİR, İRFAN; Bretting, Peter K (Springer Science and Business Media LLC, 2002-01-01)
32 Turkish corn accessions representing different climatic, geographic and topographic areas in Turkey were identified. Genetic variation and its pattern in three corn races (pop, flint and dent) included 32 accessions were investigated using 25 morphological and agronomic traits. Vegetative, ear and kernel related traits were significantly varied both among races and accessions within races. Variance component due to races and accessions within race were statistically significant, but the variance componen...
Genetic Variation in Cold Hardiness and Phenology Between and Within Turkish Red Pine (Pinus brutia Ten.) Populations: Implications for Seed Transfer
Kandemir, Gaye; Kaya, Zeki; TEMEL, FATİH; Önde, Sertaç (2010-01-01)
Wind-pollinated seeds from 40 trees (half-sib families) were collected from each of six Turkish red pine (Pinus brutia Ten.) populations in southern Turkey. Two-year old seedlings were evaluated for growth, phenology and cold resistance in a common garden experiment established in Ankara, located outside the species' natural range. Each family was represented with a six-tree row plot within each of the three replications. The below freezing temperatures (-15.2 degrees C) observed in January and February of ...
