A wild-type thermoacidophilic bacteria Bacillus acidocaldarius which excretes L-tryptophan (Trp) was used for the production of Trp. A fructose-based defined fermentation medium, i.e. containing 8 kg m(-3) fructose, 5.0 kg m(-3) (NH4)(2)HPO4, and a salt solution, was designed by investigating the influence of carbon- and nitrogen-sources and ions, to create the physiological conditions where the regulatory mechanisms in B. acidocaldarius weakly operate. The effects of oxygen transfer were investigated within limited-oxygen transfer (LimOT), low-oxygen transfer (LOT), and medium-oxygen transfer (MOT) conditions by using the parameters Q(O)/V-R =0.2 and 0.5 vvm and N=250 and 500 min(-1) in V=3 dm(3) bioreactors, at initial pH 5.0 and T=55 degrees C. The fructose, cell, Trp and the other amino acid and organic acid concentrations, the oxygen transfer rate (OTR), oxygen uptake rate (OUR), and oxygen transfer coefficients (K(L)a) were determined throughout the bioprocess; moreover, the yield and maintenance coefficients as well as the kinetic constants were reported. In general, the OTR and OUR decrease as the oxygen transfer decreases towards the limited-oxygen transfer conditions, i.e. LimOT. Damkohler number (Da) that is the maximum possible oxygen utilization rate (OD) per maximum possible oxygen-transfer rate (OTRmax) decreases with the decrease in Q(O)/V-R and/or N, that is, the oxygen-transfer resistance increases. The fructose consumption rate is the highest at MOT and the lowest at LimOT condition. In general, the cell growth rate is maximum when both the oxygen-transfer and biochemical reaction resistances are effective, i.e. at MOT. Trp production favours limited oxygen transfer conditions as B. acidocaldarius produces 0.34 kg m(-3) Trp with the highest yield at LimOT condition, where the highest total amino acid concentration was obtained as 0.36 kg m(-3), and overall Trp yield on fructose was 0.3 kg kg(-1) at t=17 h. Total amino acid concentration increased with the decrease in the oxygen transfer, despite the fact that the other aromatic group amino acids, i.e. L-tyrosine and L-phenylalanine, favour MOT condition. To open a new avenue to the production of Trp probably due to the increased reaction rates and the weakness of the regulatory mechanisms in the aromatic amino acid pathway, the fermentation characteristics of B. acidocaldarius were determined to start strain improvement research using metabolic engineering techniques. SURVIVAL KINETICS OF LACTIC-ACID STARTER CULTURES DURING AND AFTER FREEZE-DRYING
BOZOGLU, F; OZILGEN, M; BAKIR, U (Elsevier BV, 1987-09-01)
Survival kinetics of lactic acid starter cultures were modeled considering the microorganism and external medium interfacial area as the critical factors determining the resistance of the microorganisms to freeze-drying. Surviving fraction of the microorganisms increased with the increasing biomass concentration during freeze-drying, and this is attributed to the mutual shielding effect of the microorganisms against the severe conditions of the external medium. Survival of the microorganisms over the storag...
Comparison of benzaldehyde lyase production capacity in recombinant Escherichia coli and recombinant Bacillus species
Kaya, Hande; Çalık, Pınar; Department of Chemical Engineering (2006)
In this study, the benzaldehyde lyase (BAL, EC 4.1.2.38) production in E. coli BL21 (DE3) pLySs as intracellular and in Bacillus species as extracellular were investigated, and comparison of the production capacity of the enzyme in the developed recombinant microorganisms were compared. For this purpose, firstly, PCR amplified bal gene was cloned into pRSETA vector which is under the control of strong T7 promoter and expressed in E. coli BL21 (DE3) pLysS strain. With developed recombinant E. coli BL21 (DE3)...
Recombinant therapeutic protease production by Bacillus sp.
Korkmaz, Nuriye; Çalık, Pınar; Department of Chemical Engineering (2007)
The first aim of this study is the development of extracellular recombinant therapeutic protease streptokinase producing Bacillus sp., and the second aim is to determine fermentation characteristics for streptokinase production. In this context, the signal (pre-) DNA sequence of B.licheniformis (DSM1969) extracellular serine alkaline protease enzyme gene (subC: Acc. No. X03341) was ligated to 5’ end of the streptokinase gene (skc: Acc. No. S46536) by SOE (Gene Splicing by Overlap Extension) method through P...
Affinity purification and characterization of a G-protein coupled receptor, Saccharomyces cerevisiae Ste2p

We present an example of expression and purification of a biologically active G-protein coupled receptor (GPCR) from yeast. An expression vector was constructed to encode the Saccharomyces cerevisiae GPCR a-factor receptor (Ste2p, the STE2 gene product) containing a 9-amino acid sequence of rhodopsin that served as an epitope/affinity tag. In the construct, two glycosylation sites and two cysteine residues were removed to aid future structural and functional studies. The receptor was expressed in yeast cell...
Bacilysin biosynthesis by a partially-purified enzyme fraction from Bacillus subtilis
Yazgan, A; Özcengiz, Gülay; Ozcengiz, E; Kilinc, K; Marahiel, MA; Alaeddinoglu, NG (Elsevier BV, 2001-10-04)
Biosynthesis of dipeptide antibiotic bacilysin by a partially purified enzyme prepared from Bacillus subtilis PY79 was studied. Cell material was desintegrated by treatment with lysozyme and sonication and the extract was subjected to ammonium sulfate fractionation. Bacilysin-synthesizing enzyme activity was precipitated between 40% to 70% ammonium sulfate saturation. In vitro enzymatical synthesis of bacilysin was confirmed by performing thin layer chromatographic comparison of the antibiotic formed with t...
