In this work NADH-cytochrome b5 reductase enzyme (EC 1.6.2.2) obtained from rabbit liver microsomes was immobilized into gelatin by crosslinking. Chromium (III) acetate and chromium (III) sulfate were used as crosslinkers. To find the most suitable immobilization parameters; effect of crosslinker, enzyme and gelatin concentrations on immobilized enzyme activity were investigated. Unbounded enzyme was determined by washing experiments.

PRODUCTION OF SUPEROXIDE RADICALS BY SHEEP LIVER NITROFURANTOIN REDUCTASE - IN-VITRO EFFECTS OF NITROFURANTOIN METABOLITES ON DNA
ISCAN, M; MODARESI, M; Güray, Nülüfer Tülün (1995-10-01)
The reduction of nitrofurantoin by purified liver nitrofurantoin reductase was followed by the production of superoxide radicals, which were detected by the reduction of epinephrine. The conditions for the formation of superoxide radicals were optimized. The maximum superoxide radical formation occurred at approximately 3.5 mu g purified reductase, with optimum pH of 7.8 and at 0.05 mM nitrofurantoin concentration. The K-m and V-max values were calculated as 1.95 x 10(-2) mM and 4.81 nmol superoxide formed ...
Immobilization of cholesterol oxidase in a conducting copolymer of thiophene-3-yl acetic acid cholesteryl ester with pyrrole
Çırpan, Ali; Toppare, Levent Kamil; YAGCI, Y (Informa UK Limited, 2003-01-01)
Cholesterol oxidase has been immobilized in conducting copolymers of thiophene-3-yl acetic acid cholesteryl ester with pyrrole (CM/PPy) and polypyrrole (PPy) via electropolymerization. p-Toluene sulphonic acid was used as the supporting electrolyte. Kinetic parameters (V-max and K-m) and operational stability of enzyme electrodes were investigated. Surface morphology of the films was examined by scanning electron microscope.
Immobilization of glucose oxidase and urease in hydrogel matrices
Gurel, I; Arica, MY; Hasırcı, Vasıf Nejat (1997-01-01)
Immobilization of glucose oxidase and urease in hydrogels of 2-hydroxyethyl methacrylae, and N-vinyl pyrrolidone (NVP) was achieved by irradiation (using UV kand gamma-rays). The effect of radiation on entrapment efficiencies, retention of activities and swelling rates was obtained. To optimize the system, duration of exposure, reaction temperature, co-monomer concentrations, initiator and cross linker compositions were varied. The repeated reusability of the generated products was also tested. It was found...
Immobilization of glucose isomerase in surface-modified alginate gel beads
TÜMTÜRK, HAYRETTİN; DEMİREL, Gökhan; ALTINOK, HAYDAR; AKSOY, SERPİL; Hasırcı, Nesrin (2008-04-01)
In this study, glucose isomerase enzyme was entrapped into modified and nonmodified calcium alginate gel beads. Various characteristics of free and immobilized enzymes such as the optimum pH, temperature and dependence of activity on storage and operational stability were evaluated. The optimum pH and temperature of free and immobilized glucose isomerase were found to be the same values as 7.5 and 60C, respectively. For free and immobilized enzymes, kinetic parameters were calculated as 1.79 x 10(-2) and 8....
Oligomerization and cell surface expression of recombinant GABA(A) receptors tagged in the delta subunit
Oflaz, Furkan Enes; Son, Çağdaş Devrim; Arslan, Ayla (IMR Press, 2019-12-01)
The gamma-Aminobutyric acid type A receptors (GABA(A) Rs) are heteropentameric chloride channels responsible for primary inhibition in the mammalian brain. Studies have shown the expression of recombinant GABA(A) R subunits tagged with the green fluorescent protein (GFP), a 26.9 kDa protein that exhibits bright green fluorescence when exposed to light in the blue to ultraviolet range. This allows the formation of recombinant proteins essential for the development of relevant in-vitro and in-vivo methodologi...
