Comparison of RAPD-PCR and PFGE analysis for the typing of Streptococcus thermophilus strains isolated from traditional Turkish yogurts
Ghazi, Fatima; Kihal, Mebrouk; Altay, Neslihan; Gültekin, Güzin Candan (Springer Science and Business Media LLC, 2016-09-01)
The dairy industry is constantly developing new starters with specific traits. Therefore, strains from natural sources have huge potential for exploration and characterization. The aim of this work was to identify the most efficient, useful, and reliable option for typing 55 Streptococcus thermophilus isolates from traditional Turkish yogurts and four industrial strains. Then, we statistically evaluated whether our conclusions could be extended to the true population. For this purpose, the results of the ra...

A kinetic study on the plasmid stability of three Lactococcus lactis strains
Avsaroglu, M. Dilek; Buzrul, Sencer; Sanlibaba, Pinar; Alpas, Hami; Akcelik, Mustafa (Springer Science and Business Media LLC, 2007-11-01)
The plasmid stability of three wild type Lactococcus lactis strains and their mutants was investigated at different incubation time and temperatures in two different media [M17 broth and reconstituted skim milk (RSM)]. The results showed that both incubation times and temperature are effective on plasmid loss. The plasmid profiles of wild type strains exhibited 8 to 9 distinct plasmid species with molecular weights from 2.1 to 24.0 kb. Lactose fermentation ability was found to be encoded by 22.2 (strain U70...
Evaluation of structural changes induced by high hydrostatic pressure in Leuconostoc mesenteroides
Kaletunc, G; Lee, J; Alpas, Hami; Bozoglu, F (American Society for Microbiology, 2004-02-01)
Scanning electron microcopy (SEM), transmission electron microscopy (TEM), and differential scanning calorimetry (DSC) were used to evaluate structural changes in Leuconostoc mesenteroides cells as a function of high-hydrostatic-pressure treatment. This bacterium usually grows in chains of cells, which were increasingly dechained at elevated pressures. High-pressure treatments at 250 and 500 MPa also caused changes in the external surface and internal structure of cells. Dechaining and blister formation on ...
Investigation of granulation of a mixture of suspended anaerobic and aerobic cultures under alternating anaerobic/microaerobic/aerobic conditions
Bayramoğlu, Tuba Hande (Elsevier BV, 2005-12-01)
This is the first granulation study except Ferguson [Ferguson LN. Anaerobic codigestion of aircraft deicing fluid and microaerobic studies. M.S. Thesis. Milwaukee, WI, USA: Marquette University; 19991 to develop coupled granules by using a mixture of suspended anaerobic and aerobic cultures exposed to alternating cyclic anaerobic/microaerobic/aerobic conditions. Coupled granules with median sizes of 1.28-1.86 mm and settling velocities of 31-39 m/h were developed, which were comparable to those of both anae...
