Isolation of antimicrobial molecules from agricultural biomass and utilization in xylan-based biodegradable films
Çekmez, Umut; Bakır, Ufuk; Department of Biotechnology (2010)
Cotton stalk lignin extractions were performed via alkaline methods at different conditions. Crude and post treated cotton stalk lignins, olive mill wastewater and garlic stalk juice were examined in terms of antimicrobial activity. Antimicrobial lignin was isolated depending on alkaline extraction conditions. Lignin extracted at 60°C exhibited significant antimicrobial effect towards both Escherichia coli and Bacillus pumilus. However different post treatments such as ultrasonication and TiO2-assisted phot...
Isolation, characterization and immobilization of polyphenol oxidases from mulberry(morus alba) leaf tissues
Sutay, Didem; Bakır, Ufuk; Department of Chemical Engineering (2003)
In this study, the aim was to find an economical plant source for polyphenol oxidase (PPO) production as an alternative to mushroom and possible application areas by characterization and immobilization of the PPOs. For this purpose, tissues of various plants of no commercial value were screened for their PPO activities. Mulberry leaf tissues showed the highest PPO activity against 4-methyl catechol which was comparable to that of mushroom. Average Km and Vmax values of free mulberry leaf PPOs were found as ...
Isolation of volatile oil from thyme (Thymbra spicata) by steam distillation
Hanci, SS; Şahin, Serpil; Yılmaz, Levent (2003-08-01)
The effects of steam flow rate (1.03 and 0.64 L/h), distillation time, and particle size (0.50, 1.00, 2.05 mm) of Thymbra spicata on essential oil yield and sequence of extraction of compounds were studied. A logarithmic model fitted well to experimental data. The composition of the essential oil obtained by steam distillation was investigated for whole leaves only since the grinding had an adverse effect on yield. The oil obtained was very rich with respect to its carvacrol content. For both steam flow rat...
Separation of the protease enzymes of Bacillus licheniformis from the fermentation medium by crossflow ultrafiltration
Takac, S; Elmas, S; Çalık, Pınar; Ozdamar, TH (2000-06-01)
The separation from fermentation medium of extracellular serine alkaline protease (SAP) enzyme produced by Bacillus licheniformis was investigated using a crossflow ultrafiltration system. SAP was separated from the high molecular weight neutral protease (NP) and amylase (AMY) enzymes and from the low molecular weight organic acids and amino acids in a crossflow ultrafiltration system with 30000 Da and 10000 Da MWCO polysulfone membranes, respectively. The effects of transmembrane pressure (TMP), recirculat...
