The effect of cysteine-43 mutation on thermostability and kinetic properties of citrate synthase from Thermoplasma acidophilum
Kocabıyık, Semra; Russel, RJM; Danson, MJ; Hough, DW (Elsevier BV, 1996-07-05)
In this study, we have substituted serine-43 by cysteine in the recombinant citrate synthase from a moderately thermophilic Archaeon Thermoplasma acidophilum, for site-specific attachment of labels and have investigated the effects of this mutation on the biochemical properties and thermal stability of the enzyme. Both wild-type and the mutant enzymes were purified to homogenity using affinity chromatography on Matrex Gel Red A. The mutant Thermoplasma citrate synthase is very similar to wild-type citrate s...
The Effect of oxygen transfer conditions on recombinant glucose isomerase production by Pichia pastoris under glyceraldehyde-3-phosphate dehydrogenase promoter
Güneş, Hande; Çalık, Pınar; Özdamar, Tunçer H.; Department of Chemical Engineering (2015)
The aim of this study is to investigate effects of oxygen transfer conditions on recombinant glucose isomerase (r-GI) production by Pichia pastoris under glyceraldehyde-3-phosphate dehydrogenase promoter (PGAP). Two different sets of operation strategies were investigated in terms of oxygen transfer conditions. In the first one, aeration rate was kept constant at QO/V = 3 vvm, 6 vvm, and 10 vvm while agitation rate was kept at N = 900 rpm; and in the second one, dissolved oxygen concentration was kept const...
The effect of hybrid vigor on callus induction and plant regeneration from mature embryo culture of barley (Hordeum vulgare)
Ozgen, M; Birsin, MA; Onde, S (Springer Science and Business Media LLC, 2005-07-01)
The influence of hybrid vigor on callus induction, callus weight, regeneration capacity of callus, culture efficiency and number of regenerated plants from mature embryo cultures of winter barley ( Hordeum vulgare L.) hybrid was studied. A total of 15 F-1 hybrids and their parents were used for mature embryo culture. The statistical analysis of the results revealed that positive heterosis was noticed for all these characters except for callus weight. Plants regenerated in vitro were successfully established...
The effect of crosslinking on thermal inactivation of cellulases
Bilen, J; Bakir, U (1998-04-22)
Cellulase complex was crosslinked by using three different crosslinkers, dimethyl suberimidate (DMS), dimethyl adipimidate (DMA) and dimethyl-3-3'-dithiobispropionimidate (DTBP) and their effects on thermal inactivation were investigated. These reagents are all homobifunctional bisimidoesters having different crosslinking distances and crosslink only amino groups to each other. No significant activity loss was observed due to crosslinking with none of the crosslinkers used. In general, DMA and DTBP crosslin...
