Molecular cloning and co-expression of Thermoplasma volcanium proteasome subunit genes
Kocabıyık, Semra; Zwickl, Peter; Ozdogan, Seda (Elsevier BV, 2010-10-01)
In this study we describe, the construction of a co-expression vector allowing simultaneous production of Thermoplasma volcanium 20S proteasome alpha- and beta-subunits in Escherichia coli. This heterologous expression system provided high level production of fully active 205 proteasome that can be purified easily by using a conventional two-step chromatographic technique. The recombinant proteasome was purified to homogeneity 12-fold with a specific activity of 26.5 U/mg. Sodium dodecyl sulfate-polyacrylam...
Molecular mechanisms of vincristine and paclitaxel resistance in mcf-7 cell line
Demirel Kars, Meltem; Gündüz, Ufuk; Department of Biotechnology (2008)
Resistance to broad spectrum of chemotherapeutic agents in cancer cell lines and tumors has been called multiple drug resistance (MDR). In this study, the molecular mechanisms of resistance to two anticancer agents (paclitaxel and vincristine) in mammary carcinoma cell line MCF-7 were investigated. MCF-7 cells were selected in the presence of paclitaxel and vincristine by stepwise dose increments. The cell viability and growth profiles of resistant sublines were examined. As the resistance indices increased...
Molecular characterization of salmonella isolates from animals and various foods /
Durul, Bora; Soyer, Yeşim; Akkaya, Mahinur S.; Department of Biotechnology (2014)
Salmonella, which is a Gram-negative human and animal pathogen, poses a significant threat to public health with its existence and survival in the farm to fork chain. Of the greater importance, Salmonella enterica subsp. enterica, which covers 60 % of nearly 2500 Salmonella serotypes, causes diseases in mammals with varying host affiliations and pathogenity levels. In order that consumer safety can be assured, elimination of this harmful pathogen from the farm to fork chain is a must, thereby bringing the i...
Molecular adaptations in cadmium and lead resistant environmental species
Kepenek, Eda Şeyma; Gözen, Ayşe Gül; Severcan, Feride; Department of Biology (2017)
Environmental pollution caused by heavy metal exposure has detrimental effects on human health. For the reclamation of polluted areas, bacteria have been used in remediation. For those bioremediation attempts, finding the appropriate bacterial strains is an important issue. In their environment, bacteria may exist close to the source of heavy metals. Sudden exposure to high concentrations of the heavy metal may trigger quick response mechanisms to ensure survival. On the other hand, bacteria may live at a d...
