Partial purification and characterization of neutral trehalase from commercial baker's yeast, Saccharomyces cerevisiae
Yarar, S; Hamamcı, Haluk; Bakir, U (2000-12-01)
The neutral trehalase of a commercial baker's yeast (S. cerevisiae) strain has been partially purified using ammonium sulfate fractionation and DEAE-cellulose column chromatography techniques. Trehalase was precipitated between 35-50% ammonium sulfate saturation and approximately 5-8 fold purification was achieved. The yeast cAMP-dependent protein kinase was also precipitated in the same fraction and these two proteins were separated by DEAE-cellulose column chromatography. Trehalase became totally inactive...
Evaluating microemulsions for purification of beta-galactosidase from Kluyveromyces lactis
Mazı, Bekir Gökçen; Hamamcı, Haluk; Department of Food Engineering (2010)
In this study, we evaluated the potential of water-in-oil microemulsions for the separation of beta-galactosidase (lactase) from other proteins. The ability of beta-galactosidase to break down the milk carbohydrate lactose gives the enzyme considerable commercial importance. The extent of solubilization of a commercial Kluyveromyces lactis preparation of beta-galactosidase into microemulsion droplets formed from 200 mM bis (2-ethylhexyl) sodium sulfosuccinate (AOT) in isooctane was measured as a function of...
Prediction of protein subcellular localization based on primary sequence data
Özarar, Mert; Atalay, Mehmet Volkan; Department of Computer Engineering (2003)
Subcellular localization is crucial for determining the functions of proteins. A system called prediction of protein subcellular localization (P2SL) that predicts the subcellular localization of proteins in eukaryotic organisms based on the amino acid content of primary sequences using amino acid order is designed. The approach for prediction is to nd the most frequent motifs for each protein in a given class based on clustering via self organizing maps and then to use these most frequent motifs as features...
The effect of crosslinking on thermal inactivation of cellulases
Bilen, J; Bakir, U (1998-04-22)
Cellulase complex was crosslinked by using three different crosslinkers, dimethyl suberimidate (DMS), dimethyl adipimidate (DMA) and dimethyl-3-3'-dithiobispropionimidate (DTBP) and their effects on thermal inactivation were investigated. These reagents are all homobifunctional bisimidoesters having different crosslinking distances and crosslink only amino groups to each other. No significant activity loss was observed due to crosslinking with none of the crosslinkers used. In general, DMA and DTBP crosslin...
Account Books of the Imperial WAQFS Charitable Endowments in the Eastern Mediterranean 15th to 19th Centuries
Orbay, Kayhan (American Accounting Association, 2013-01-01)
The history of accounting in the Eastern Mediterranean has not been adequately studied through its primary sources, despite the fact that the Turkish archives house an enormous amount of material for exploring accounting practices in the Ottoman Empire. Ottomanists used the account books as sources for Ottoman socioeconomic and institutional history. They analyzed, fully transliterated and published the account registers of the central treasury, the Istanbul shipyard and the waqf institutions. Nevertheless,...
