Carbapenem resistance in Acinetobacter baumannii is a growing public health concern and represents a serious problem for treatment of the infection. Several carbapenem-hydrolysing β-lactamases have been identified from A. baumannii so far. In this study carbapenem resistance in A.baumannii strains recovered from intensive care units of Gulhane Military Medical Academy, Turkey, were investigated via multiplex PCR and with parallel phenotypic tests. From June 2006 to January 2010, 138 clinical A. baumannii isolates were collected. Identification and antimicrobial susceptibility tests of the isolates were performed. The MICs of imipenem and meropenem were determined by using E-test method. Carbapenem resistant A. baumannii strains were included for further study. Firstly, the presence of carbapenemases were determined. The presence of Metallo-beta-lactamase (MBL) were also investigated. Detection of the four groups of OXA carbapenemases (OXA- 23, OXA-24, OXA-51 and OXA-58) was carried out using a multiplex PCR assay. Sequence analyses were performed. Non-duplicate, multidrug resistant 61 clinical A. baumannii isolates were found to be resistant to imipenem and meropenem. In the 61 isolates, the MIC50 of imipenem and meropenem were 16 and >32; MIC90 were 192 and >32 respectively. Modified Hodge Tests (MHT) were positive for all 61 A. baumannii strains. None of these isolates showed MBL activity. As determined through multiplex PCR, all of the 61 isolates had blaOXA-51 genes, 50 isolates had blaOXA-23, and 11 isolated had blaOXA-58 genes. Alleles encoding OXA-24-like enzymes were not detected in any isolates. This study indicated that the clinical isolates in our region contained blaOXA-51, blaOXA-23, and blaOXA-58 resistance genes. However, blaOXA-24 gene was either absent or occur in very low frequency.

Evaluation of colloidal platinum on cytotoxicity, oxidative stress and barrier permeability across the gut epithelium

Colloidal platinum (Pt) is widely consumed due to its health promoting benefits. However, the exact biological effects of these nanoparticles have not been studied in detail, particularly in the gut. In the present study we observed that colloidal Pt was not cytotoxic towards three different epithelial colon cancer cell lines. Co-treatment of the colon cancer cell line Caco-2 with the oxidative stress inducing agent hydrogen peroxide (H2O2) and colloidal Pt resulted in a significant decrease in H2O2 induced...
Molecular investigation of ptz-induced epileptic activities in rat brain cell membranes and the effects of vigabatrin
Görgülü Türker, Sevgi; Severcan, Feride; Department of Biology (2009)
The epilepsies are a heterogenous group of symptom complexes, whose common features is the recurrence of seizures. There is no certain therapy for epilepsy. In order to promote new advances for the prevention of epilepsy the molecular mechanism of epileptic activities should be clarified. In the present study the goal is to obtain information for molecular mechanism of epilepsy. To achieve this, molecular alterations from pentylenetetrazol (PTZ)-induced epileptic activities on rat brain tissue and cell memb...
Determination of colistin resistance in Escherichia coli isolates from foods in Turkey, 2011-2015
Güzel, Mustafa; Avşaroğlu, M Dilek; Soyer, Yeşim (2020-05-01)
Antimicrobial resistance of pathogenic microorganisms is an emerging public health concern. Intensive use of antibiotics in food animals might increase antimicrobial resistance in foodborne pathogens. Colistin is a last resort antibiotic for treatment of multidrug resistant (MDR) Gram negative pathogens. The recent antimicrobial resistance studies revealed a mobile antimicrobial resistance gene (mcr) that provides resistance to colistin. Furthermore, the gene has been found in different genera. Therefore, t...
Preparation of polyethylene glycol coated magnetic nanoparticles for targeting of cancer cells
Keskin, Tuğba; Gündüz, Ufuk; Department of Biology (2012)
Conventional cancer chemotherapies cannot differentiate between healthy and cancer cells, and lead to severe side effects and systemic toxicity. In the last decades, different kinds of controlled drug delivery systems have been developed to overcome these shortcomings of chemotherapeutics. Magnetic nanoparticles (MNP) are potentially important in cancer treatment since they can be targeted to tumor site by an externally applied magnetic field. In this study, it is aimed to synthesize folic acid conjugated; ...
Cytotaxonomic studies on the genus salvia (labiatae) in Turkey
İnanç Gök, Tuğba; Doğan, Musa; Department of Biology (2009)
The genus Salvia L. is significantly important with regard to both its worldwide distribution and usage areas including food, medical and perfumary industries. In this current study, it is targeted to address the chromosome numbers and karyomorphology of the ten species and one variety of the genus Salvia. All of the eleven taxa examined in this study are economically significant and nine of these are endemic to Turkey. To define the chromosome numbers and karyomorphology of these eleven taxa somatic chromo...
