The objective of this study was to develop and evaluate the anticancer activity and the safety of a combinational drug delivery system using polyamidoamine (PAMAM) dendrimer-coated iron oxide nanoparticles for doxorubicin and poly I:C delivery in vitro. Dendrimer-coated magnetic nanoparticles (DcMNPs) are suitable for drug delivery system as nanocarriers with their following properties, such as surface functional groups, symmetry perfection, internal cavities, nano-size and magnetization. These nanoparticles could be targeted to the tumor site under a magnetic field since they have a magnetic core. DcMNPs were found as a convenient vehicle for targeted doxorubicin delivery in cancer therapy. Poly (I:C) binding on doxorubicin loaded DcMNPs (DcMNPs-Dox) was reported for the first time in the literature. It was also demonstrated that loading of doxorubicin into the cavities of DcMNPs increases the binding efficiency of poly (I:C) to the surface functional groups of dendrimer up to 10 times. When we compare the in vitro cytotoxic properties of doxorubicin, poly (I:C) and poly (I:C) bound doxorubicin loaded DcMNPs (PIC-DcMNPs-Dox), it was observed that PIC-DcMNPs-Dox show the highest cytotoxic effect by passing the cell resistance mechanisms on doxorubicin resistant MCF7 (MCF7/Dox) cells. Results demonstrated that applying PIC-DcMNPs-Dox would improve the efficacy by increasing the biocompatibility of system in blood stream and the toxicity inside tumor cells. These results provide invaluable information and new insight for the design and optimization of a novel combinational drug delivery system for targeted cancer therapy. (C) 2014 Elsevier Masson SAS. Chitosan superparamagnetic nanoparticles, loaded with Doxorubicin and Bortezomib were synthesized for treatment of breast and cervical cancers by targeted drug delivery. In vitro cytotoxicity analyses revealed that the efficacy of drugs was highly increased when applied as loaded on nanoparticles. Chitosan superparamagnetic iron oxide nanoparticles (CSMNPs) were in-situ synthesized at different sizes by ionic crosslinking method. The characterization of nanoparticles was performed by XRD, XPS/ESCA, FTIR, TE...

In our effort for the development of novel anticancer therapeutics, a series of isoxazole-piperazine analogues were prepared, and primarily screened for their antiproliferative potential against hepatocellular carcinoma (HCC; Huh7/Mahlavu) and breast (MCF-7) cancer cells. All compounds demonstrated potent to moderate cytotoxicity on all cell lines with IC50 values in the range of 0.09-11.7 mu M. Further biological studies with 6a and 13d in HCC cells have shown that both compounds induced G1 or G2/M arrests...
The Antiproliferative Effect of Celecoxib Loaded pNIPAM Nanoparticles

The aim of this study was to design a drug delivery system based on poly(N-isopropylacrylamide) (pNIPAM) nanoparticles (NPs). The model drug, Celecoxib, is a cyclooxygenase-2 inhibitor and has a great potential in chemoprevention and treatment of various cancer types, however, the clinical use is limited due to the side effects on the cardiovascular system which is most probably due to the high doses used in clinical applications. In this study, a novel nanoparticle preparation approach, nanoprecipitation, ...

Analysis of the genetic determinant for production of the pediocin P of Pediococcus pentosaecus Pep 1
Osmanagaoglu, O; Beyatli, Y; Gündüz, Ufuk; Sacilik, SC (2000-01-01)
Pediococcus pentosaceus Pep1 is a vacuum-packaged Turkish sausage isolate which produces a potentially novel bacteriocin of the pediocin (anti-Listeria) family of peptides designated as pediocin P. Curing experiments and plasmid profile analysis indicated that both bacteriocin immunity and production determinants were linked and encoded by 9.0 MDa plasmid, pHD1.0. Attempts to transform purified plasmid pHD1.0 into recipient Escherichia coli JM109 cells by electroporation were successful but none of the E. c...
