Batch-wise sorbitol addition as a co-substrate at the induction phase of methanol fed-batch fermentation by Pichia pastoris (Mut(+)) was proposed as a beneficial recombinant protein production strategy and the metabolic responses to methanol feeding rate in the presence of sorbitol was systematically investigated. Adding sorbitol batch-wise to the medium provided the following advantages over growth on methanol alone: (a) eliminating the long lag-phase for the cells and reaching 'high cell density production' at t = 24 h of the process (C-X = 70 g CDW/I); (b) achieving 1.8-fold higher recombinant human erythropoietin (rHuEPO) (at t = 18 h); (c) reducing specific protease production 1.2-fold; (d) eliminating the lactic acid build-up period; (e) lowering the oxygen uptake rate two-fold; and (f) obtaining 1.4-fold higher overall yield coefficients. The maximum specific alcohol oxidase activity was not affected in the presence of sorbitol, and it was observed that sorbitol and methanol were utilized simultaneously. Thus, in the presence of sorbitol, 130 mg/l rHuEPO was produced at I = 24 h, compared to 80 mg/l rHuEPO (t = 24 h) on methanol alone. This work demonstrates not only the ease and efficiency of incorporating sorbitol to fermentations by Mut(+) strains of P. pastoris for the production of any bio-product, but also provides new insights into the metabolism of the methylotrophic yeast P. pastoris. Oxygen transfer as a tool for fine-tuning recombinant protein production by Pichia pastoris under glyceraldehyde-3-phosphate dehydrogenase promoter
Gunes, Hande; Çalık, Pınar (2016-07-01)
Effects of oxygen transfer on recombinant protein production by Pichia pastoris under glyceraldehyde- 3-phosphate dehydrogenase promoter were investigated. Recombinant glucose isomerase was chosen as the model protein. Two groups of oxygen transfer strategies were applied, one of which was based on constant oxygen transfer rate where aeration rate was Q(O)/V = 3 and 10 vvm, and agitation rate was N = 900 min(-1); while the other one was based on constant dissolved oxygen concentrations, C-DO = 5, 10, 15, 20...
Fed-batch cultivation of bakers' yeast: Effect of nutrient depletion and heat stress on cell composition
Ertugay, N; Hamamci, H; Bayındırlı, Alev (Springer Science and Business Media LLC, 1997-01-01)
The physiology of a commercial strain of bakers' yeast was studied in terms of the cell composition under different growth conditions and of its response to stress. The study comprised fed-batch experiments since this is the system used in bakers' yeast industry. The classical fed-batch fermentation procedure was modified in that the yeast cells were continuously grown to a steady-state at a dilution rate of 0.1/h in order to achieve more or less the same initial starting point in terms of cell composition....
High hydrostatic pressure-assisted extraction of lipids from Lipomyces starkeyi biomass
Tuhanioglu, Arda; Alpas, Hami; Çekmecelioğlu, Deniz (2022-10-01)
The purpose of this study is to evaluate the effect of high hydrostatic pressure (HHP) as a novel approach for yeast cell disruption and lipid extraction from Lipomyces starkeyi DSM 70295 grown in glucose medium (40 g/L and C/N:55/1) at initial pH of 5.0, 25 degrees C, and 130 rpm for 8 days. HHP extraction conditions including pressure, time, and temperature were optimized by response surface methodology. The high speed homogenizer-assisted extraction (HSH) was also used for comparison. The biomass subject...
Anaerobic acidification of sugar-beet processing wastes: Effect of operational parameters
Alkaya, Emrah; Demirer, Göksel Niyazi (Elsevier BV, 2011-01-01)
The objective of this study was to maximize the hydrolysis and acidification of sugar-beet processing wastewater and beet pulp for volatile fatty acid (VFA) production through acidogenic anaerobic metabolism. Experiments were conducted to determine the optimum operational conditions (HRT, waste-mixing ratio and pH) for effective acidification in daily-fed, continuously mixed anaerobic reactors. For this purpose, reactors were operated at 35 +/- 1 degrees C with different combinations of HRT (2-4 days), wast...
Co-substrate mannitol feeding strategy design in semi-batch production of recombinant human erythropoietin production by Pichia pastoris
Eskitoros, Melda S.; Çalık, Pınar (2014-05-01)
BACKGROUND The effects of alternative co-substrate feeding strategies on recombinant human erythropoietin (rHuEPO) production by Pichia pastoris-Mut(+) strain were investigated. RESULTS Five different production strategies were designed and performed in the production phase of the bioprocesses with the use of either mannitol or sorbitol as the co-substrate. The highest rHuEPO production was achieved as C-rHuEPO= 0.65 g L-1 at t=9 h, with the cell concentration C-x=55 g L-1 in the production phase; wherein m...
