We have previously shown that ethanolic extract from bark (EEB) of Salix aegyptiaca (Musk Willow) can inhibit proliferation and motility and induce apoptosis in colon cancer cells. Tandem mass spectrometry revealed EEB to be rich in catechin, catechol, and salicin. The present study investigated the chemopreventive effect of HPLC-fingerprinted EEB on 1,2-dimethylhydrazine (DMH)-induced aberrant crypt foci (ACF) formation in mice. DMH (20mg/kg body weight) was weekly injected subcutaneously to mice for the first 2weeks. EEB (100 and 400mg/kg body weight) was provided orally from the 7th to 14th week, after which colon tissues were evaluated histologically and biochemically. DMH treatment induced high number of ACF; EEB significantly reduced the number and multiplicity of ACF, along with a restoration in goblet cells and mucin accumulation. EEB supplementation improved the markers of inflammation (myeloperoxidase and neutrophil infiltration) and oxidative stress. More importantly, EEB amplified apoptosis of neoplastic cells in the colon mucosa of DMH-treated mice. It also lowered levels of markers for early transformation events such as EGFR, nuclear -catenin, and COX-2 in colon cancer cell lines HT-29 and HCT-116. The innocuity of EEB (up to 1600mg/kg) to mice reinforces its potential as a chemopreventive agent.

Antioxidant capacities and bioactive compounds of novel thermo-tolerant green microalgae
Bulut, Onur; Öktem, Hüseyin Avni; Department of Biotechnology (2018)
Accumulation of reactive oxygen species (ROS) at high levels in cells as a result of normal cellular activities as well as exposure to oxidative stress is attributed to many chronic degenerative diseases and conditions in humans including cardiovascular diseases, cancer and aging-related conditions such as Alzheimer's disease. Although living organisms are equipped with enzymatic and non-enzymatic antioxidant defense systems to counteract the adverse effects of oxidative stress, yet they rely on exogenous s...
Immunotherapeutic applications of CpG-containing oligodeoxynucleotides

Bacterial DNA and synthetic oligodeoxynucleotides (ODN) expressing unmethylated CpG motifs stimulate the mammalian immune system to mount a rapid innate immune response. This response is characterized by the production of polyreactive IgM, immunomodulatory cytokines and chemokines. CpG ODN directly stimulate lymphocytes, natural killer cells and professional antigen-presenting cells (such as macrophages and dendritic cells). Owing to the strength and nature of this stimulation, CpG ODN are being harnessed f...
Immunotherapeutic utility of stimulatory and suppressive oligodeoxynucleotides.

Bacterial DNA contains immunostimulatory CpG motifs that interact with toll-like receptor 9 on immune cells to stimulate the production of cytokines, chemokines and immunoglobulins. Synthetic oligodeoxynucleotides (ODNs) containing CpG motifs mimic the activity of bacterial DNA. Recently, several structurally distinct types of CpG ODN were identified that differentially activate human immune cells. These ODNs may be useful as vaccine adjuvants, anti-allergens and in the treatment of infectious diseases and ...
Fullerol based enzyme mimics and synthesis effort of tetra aryl pyranone
Soydan, Medine; Özçubukçu, Salih; Department of Chemistry (2021-2-12)
Although, natural enzymes have high catalytic activity and specificity, they are fragile and costly due to long purification processes. Therefore, scientists started to design and synthesize structures with enzyme-like activity called nanozymes to overcome the limitations of the natural enzymes. In the first part of this project, amino acids of catalytic site of three enzymes were conjugated into fullerol. The names of three enzymes are lactate dehydrogenase, protease and nitrilase. Then, t...
Inference of the stochastic MAPK pathway by modified diffusion bridge method
Purutçuoğlu Gazi, Vilda (2013-03-01)
The MAPK pathway is one of the well-known systems in oncogene researches of eukaryotes due to its important role in cell life. In this study, we perform the parameter estimation of a realistic MAPK system by using western blotting data. In inference, we use the modified diffusion bridge algorithm with data augmentation technique by modelling the realistically complex system via the Euler-Maruyama approximation. This approximation, which is the discretized version of the diffusion model, can be seen as an al...
