Alginate is a natural biopolymer composed of mannuronic and guluronic acids. In this study, alginate was produced by Azotobacter vinelandii ATCC® 9046 in a laboratory fermenter under controlled conditions. Effect of variation of sucrose concentration on alginate quality and quantity was investigated. It appeared that 20 g/L of sucrose concentration was the optimum dose for alginate production with maximum alginate concentration of 4.5 g/L. Similarly, a higher degree of polymerization seemed to be achieved at 20 g/L of sucrose concentration. Monomer distribution of alginate was also affected by variation of sucrose concentrations. Alginate synthesized at 10 g/L of sucrose was observed to be mainly composed of guluronic acid which increased up to 89% of GG-blocks. On the other hand, almost equal amount of mannuronic and guluronic acids was obtained at 20 g/L of sucrose concentration. Higher GG-blocks could give advantage to the bacteria under unfavourable environmental conditions such as the one which opposed at low sucrose concentration. Therefore, it is possible to produce alginate with different block distributions by manipulating the concentrations of the carbon source.
