The effect of salt stress (100 mM and 200 mM NaCl) on antioxidant responses in shoots and roots of 14-day-old lentil (Lens culinaris M.) seedlings was investigated. Salt stress caused a significant decrease in length, wet-dry weight and an increase in proline content of both shoot and root tissues. In leaf tissues, high salinity treatment resulted in a 4.4 fold increase in H2O2 content which was accompanied by a significant level of lipid peroxidation and an increase in electrolyte leakage. Root tissues were less affected with respect to these parameters. Leaf tissue extracts exhibited four activity bands, of which two were identified as Cu/Zn-SOD and others as Fe-SOD and Mn-SOD. Fe-SOD activity was missing in root extracts. In both tissues Cu/Zn-SOD activity comprised 70-75% of total SOD activity. Salt stress did not cause a significant increase in total SOD activity of leaf tissues but a significant enhancement (88%) was observed in roots mainly due to an enhancement in Cu/ZnSOD isoforms. Compared to leaf tissues a significantly higher constitutive ascorbate peroxidase (APX) and glutathion reductase (GR) activity was observed in root tissues. Upon salt stress no significant change in the activity of APX, catalase (CAT) and GR was observed in root tissues but a higher APX activity was present when compared to leaf tissues. On the other hand, in leaf tissues, with the exception of CAT, salt stress caused significant enhancement in the activity of other antioxidant enzymes. These results suggested that, root tissues of lentil are protected better from NaCl stress induced oxidative damage due to enhanced total SOD activity together with a higher level of APX activity under salinity stress. To our knowledge this is the first report describing antioxidant enzyme activities in lentil.

The effects of cold and drought stress on antioxidant responses and growth parameters in shoots and roots of lentil (Lens culinaris M cv Sultan 1) seedlings were investigated. Ten-day-old hydroponically grown seedlings were subjected to drought and cold (4 degrees C) stress for 5 days. The length and fresh weight of shoots decreased significantly under both stress conditions, contrary to the increase in these growth parameters for roots under the same conditions. The oxidative damage as:generation of malond...

In this study, the effects of long-term NaCl treatment were investigated in two cultivars of peanut designated as drought-resistant and drought-sensitive. Growth parameters, changes in the concentrations of MDA, H2O2 and proline, and the activities of antioxidant enzymes were determined under salinity stress. Growth parameters indicated the superiority of cv. Florispan to cv. Gazipasa under milder salinity stress treatment. However, comparative analysis of the two cultivars showed that MDA, H2O2, ion leakag...
Antioxidant responses of tolerant and sensitive barley cultivars to boron toxicity
Karabal, E; Yucel, M; Öktem, Hüseyin Avni (2003-06-01)
Effect of boron toxicity on antioxidant response of sensitive (Hamidiye) and tolerant (Anadolu) barley (Holgarium vulgar) cultivars was investigated. Eight days old hydroponically grown seedlings were subjected to 5 and 10 mM boric acid treatments for 5 days. Growth parameters (dry-wet weight), protein, proline, MDA, H2O2 contents, membrane damage and activities of superoxide dismutase (SOD). ascorbate peroxidase (APX), catalase (CAT) and glutathion reductase (GR) was evaluated in root and shoot tissues. Co...
Production of neutral and alkaline extracellular proteases by the thermophilic fungus, Scytalidium thermophilum, grown on microcrystalline cellulose
Ifrij, IH; Ögel, Zümrüt Begüm (Springer Science and Business Media LLC, 2002-07-01)
Extracellular proteases produced by Scytalidium thermophilum, grown on microcrystalline cellulose, were most active at pH 6.5-8 and 37-45 degreesC when incubated for 60 min. Highest protease activity was at day 3 where endoglucanase activity was low. Protease activity measurements with and without the protease inhibitors, p-chloromercuribenzoate, PMSF, antipain, E-64, EDTA and pepstatin A, suggest production of thiol-containing serine protease and serine proteases. Endoglucanase and Avicel-adsorbable endogl...
Optimization of a regeneration and transformation system for lentil (lens culinaris M., cv. Sultan-I) cotyledonary petioles and epicotyls
Bayraç, Abdullah Tahir; Öktem, Hüseyin Avni; Department of Biotechnology (2004)
In this study, optimization of a transformation and regeneration system via indirect organogenesis in cotyledonary petiole tissue of lentil (Lens culinaris Medik.) was investigated. Eight different medium types differing in their plant growth regulator compositions were employed to examine the callus induction potency of cotyledonary petiole. Except two, all other tested medium yielded more than 80% callus induction. Nine different medium types were studied to test the potencies of callus structures for sho...
