1. Reductase was purified to electrophoretic homogeneity from sheep liver and lung microsomes. The specific activity of both enzymes ranged from 55 to 66 μ mol cytochrome c reduced/min/mg protein. 2. Liver and lung reductases appeared to have similar kinetic and spectral properties. Km, (NADPH) and Km (cytochrome c) values were calculated to be 14.3 ± 1.23μM and 22.2 ± 2.78 μM for liver and 11.1 ±0.70 μM and 20.0 ± 2.15μM for lung reductase, respectively. Kinetic studies showed that cytoehrome c can bind the oxidized form of the enzyme as well as its reduced form and both reductases operated through a ping-pong type mechanism. 3. These reductases cannot be distinguished on the basis of monomer molecular weights (Mr 78,000) except that the liver reductase was found to be more susceptible to proteolytic attack. 4. Both reductases supported aniline 4-hydroxylation and ethylmorphine N-demethylation reactions to the same extent in the reconstituted systems. However, sheep lung reductase appeared only 36.5 and 14.8% as effective in catalyzing benzo[a]pyrene reaction as an equivalent amount of reductase from liver in the presence of liver cytochrome P-450 and 3MC-treated rat liver cytochrome P-448, respectively.

Simultaneous purification and characterization of cytochrome b5 reductase and cytochrome b5 from sheep liver
Arinc, E; Cakir, D (Elsevier BV, 1999-02-01)
Cytochrome b5 was purified from detergent solubilized sheep liver microsomes by using three successive DEAE-cellulose, and Sephadex G-100 column chromatographies. It was purified 54-fold and the yield was 23.5% with respect to microsomes. The apparent Mr of cytochrome b5 was estimated to be 16,200 +/- 500 by SDS-PAGE. Absolute absorption spectrum of the purified cytochrome b5 showed maximal absorption at 412 nm and dithionite-reduced cytochrome b5 gave peaks at 557, 526.5 and 423 nm. The ability of the puri...
STIMULATORY EFFECTS OF LUNG CYTOCHROME B5 ON BENZPHETAMINE N-DEMETHYLATION IN A RECONSTITUTED SYSTEM CONTAINING LUNG CYTOCHROME P450LGM2
ARINC, E; PASHA, RPK; Adalı, Orhan; BASARAN, N (Elsevier BV, 1994-08-01)
1. Cytochrome b 5 was partially purified from sheep lung microsomes in the presence of detergents Emulgen 913 and cholate by three consecutive DEAE-cellulose and Sephadex G-100 gel filtration chromatographies.
COMPARATIVE-STUDIES OF SHEEP LUNG AND LIVER NITROFURANTOIN REDUCTASE
İşcan, Mesude (Elsevier BV, 1993-10-01)
1. Nitrofurantoin reductase which catalyzes the bioactivation of nitrofurantoin was purified to electrophoretic homogenity from sheep liver and lung microsomes, with a yield of 15% and 35%, respectively. The specific activity of both reductases was found to be similar (140 nmol/min/mg protein). 2. The effects of nitrofurantoin and NADPH concentrations, pH, ionic strength, amount of enzyme and reaction period, on the enzyme activity were studied and the optimum conditions for maximum activity of purified fi...
POSSIBLE INVOLVEMENT OF MANGANESE IN THE CATALYTIC MECHANISM OF BOVINE LIVER ARGINASE
TURKOGLU, S; OZER, I (Elsevier BV, 1992-06-01)
1. Bovine liver arginase followed Michaelis-Menten kinetics in the pH range of 4.5-9.0. The variation of upsilon(i) pH implied that a basic group (pK(alpha) 8.7) functions at the catalytic site.
Biochemical characteristics of purified beef liver NADPH-cytochrome P450 reductase
Arinc, E; Celik, H (Wiley, 2002-01-01)
NADPH-cytochrome P450 reductase, an obligatory component of the cytochrome P450 dependent monooxygenase system, was purified to electrophoretic homogeneity from beef liver microsomes. The purification procedure involved the ion exchange chromatography of the detergent-solubilized microsomes on first and second DEAE-cellulose columns, followed by 2',5'-ADP Sepharose affinity chromatography. Further concentration of the enzyme and removal of Emulgen 913 and 2'-AMP were accomplished on the final hydroxylapatit...
