Cloning and expression ofbenzaldehydelyase from Pseudomonas fluorescens Biovar I in Pichia pastoris
Büyüksungur, Arda; Çalık, Pınar; Department of Biotechnology (2006)
Benzaldehyde lyase (BAL, EC 4.1.2.38) from Pseudomonas fluorescens Biovar I, a thiamine pyrophosphate (ThDP) dependent enzyme, catalyzes the enzymatic kinetic resolution of racemates by C-C bond cleavage and concomitant C-C bond formation. In this study, benzaldehyde lyase gene from Pseudomonas fluorescens Biovar I was cloned into Pichia pastoris, with the aim of the extracellular production of the enzyme. For this purpose, firstly, PCR amplified bal gene was cloned into an integration vector pPICZalphaA. T...
Cloning and heterologous expression of chlorophyll a synthase in Rhodobacter sphaeroides
Ipekoglu, Emre M.; Gocmen, Koray; Oz, Mehmet T.; Gurgan, Muazzez; Yucel, Meral (2017-03-01)
Rhodobacter sphaeroides is a purple non-sulfur bacterium which photoheterotrophically produces hydrogen from organic acids under anaerobic conditions. A gene coding for putative chlorophyll a synthase (chlG) from cyanobacterium Prochlorococcus marinus was amplified by nested polymerase chain reaction and cloned into an inducible-expression plasmid which was subsequently transferred to R. sphaeroides for heterologous expression. Induced expression of chlG in R. sphaeroides led to changes in light absorption ...

Cloning and expression of trehalose-6-phosphate synthase 1 from Rhizopus oryzae
Uyar, Ebru Ozer; Yucel, Meral; Hamamcı, Haluk (2016-05-01)
Trehalose is a reducing disaccharide acting as a protectant against environmental stresses in many organisms. In fungi, Trehalose-6-phosphate synthase 1 (TPS1) plays a key role in the biosynthesis of trehalose. In this study, a full-length cDNA from Rhizopus oryzae encoding TPS1 (designated as RoTPS1) was isolated. The RoTPS1 cDNA is composed of 2505 nucleotides and encodes a protein of 834 amino acids with a molecular mass of 97.8kDa. The amino acid sequence of RoTPS1 has a relatively high homology with th...
