Poly(2,5-di(furan-2-yl)thiazolo[5,4-d]thiazol) polymer was coated over a platinum electrode and new enzyme electrodeswere produced that are not found in the literature. The formation of polymerization was shown by cyclic voltammetry.Kinetic parameters of immobilized polyphenol oxidase enzyme were determined. Vmax and Km were obtained as 0.028 ±0.001 umol min1- electrode1- and 669.68 ± 64.73 mM respectively. The effect of reaction conditions over enzyme activitywere examined. The pH and temperature at which immobilized polyphenol oxidase enzyme showed the highest activitywere found as pH 7.5 and 45 ℃. Amount of enzyme and gluteraldehyde immobilized to the electrodes were changed and inthe examination, it was detected that it is sufficient to add 2x6 uL enzyme and 1x6 uL gluteraldehyde to the electrode. Theactivity of Pt/PTTzFr/PPO electrode was followed by 50 consequential measurements. It has been seen after 50measurements that immobilized enzyme activity is 70%. As a result of shelf life examinations, it was determined thatPt/PTTzFr/PPO electrode lost 30% of its initial activity at the end of the 50th day and the remaining activity was 70%.Calibration graph was plotted for Pt/PTTzFr/PPO electrode (y = 0.0139x + 0.0479). LOQ value was calculated as 7.827 mgmL-1 and the working interval was found as 1.0 – 90.0 mg mL1-. In total phenolic material analyses made with thiselectrode, the results found in waste water samples have also been confirmed by Folin-Ciocalteau method.

Production and biochemical characterization of polyphenol oxidase from thermomyces lanuginosus

Polyphenol oxidases are enzymes that catalyze the oxidation of certain phenolic substrates to quinones in the presence of molecular oxygen. Polyphenol oxidases are widely used in several applications. In food industry, they are used for enhancement of flavor in coffee, tea and cocoa production, and determination of food quality. In medicine, they have several uses in treatments of Parkinson̕s disease, phenlyketonurea and leukemia. In wastewater treatment, they are used for the removal of phenolic pollutants...
Tailoring the optoelectronic properties of donor-acceptor-donor type pi-conjugated polymers via incorporating different electron-acceptor moieties
Tarkuc, Simge; Udum, Yasemin Arslan; Toppare, Levent Kamil (2012-01-31)
Syntheses of donor-acceptor-donor type of pi-conjugated monomers were performed to examine the effect of the acceptor units' strength on the electrochemical and optoelectrochemical properties of the resulting monomer and polymer. Palladium catalyzed Stille cross-coupling reaction of an organotin reagent with an organic electrophile was used for the synthesis of target monomers, 5,8-bis(4-hexylthiophen-2-yl)-2-(2,3-dihydrobenzo[b][1,4]clioxin-6-y1)-3-(2,3-dihydrobenzo[b][1,41dioxin-7-yl)quinoxaline (DBQHT) a...
Electrochemical polymerization of 1-(4-nitrophenyl)-2,5-di(2-thienyl)-1 H-pyrrole as a novel immobilization platform for microbial sensing

Two types of bacterial biosensor were constructed by immobilization of Gluconobacter oxydans and Pseudomonas fluorescens cells on graphite electrodes modified with the conducting polymer; poly(1-(4-nitrophenyl)-2,5-di(2-thienyl)-1 H-pyrrole) [SNS(NO2)]. The measurement was based on the respiratory activity of cells estimated by the oxygen consumption at -0.7 V due to the metabolic activity in the presence of substrate. As well as analytical characterization, the linear detection ranges, effects of electropo...
Electrochemical copolymerization of thiophene containing pseudo-polyether cages with pyrrole
Cihaner, Atilla; Önal, Ahmet Muhtar (2006-01-01)
Conducting copolymers were synthesized via the electrochemical oxidation of pyrrole (Py) in the presence of the monomer 1,12-bis(2-thienyl)-2,5,8,11-tetraoxadodecane (1). The presence of monomer I in the electrolytic solution greatly changed the CV behavior of Py during its potensiodynamic polymerization. The electroactivity of poly(I-co-Py) increased with the increasing amount of I in the comonomer mixture. Copolymer films were prepared via constant potential electrolysis in an electrolytic solution contai...
Activation of poly(dimer acid-co-alkyl polyamine) particles for covalent immobilization of alpha-amylase
Hasırcı, Nesrin; Tumturk, Hayrettin (2006-12-01)
Poly(dimer acid-co-alkyl polyamine) particles were activated by using various chemicals such as carbodiimide (CDI), ethylene diamine (EDA), and hexamethylene diamine (HMDA) and studied as support materials for covalent immobilization of alpha-amylase. The activities of the immobilized enzymes on these activated particles were investigated. The highest activity of free enzyme was obtained at pH 7.5 while this value was shifted to pH 6.5 for CDI and EDA activated systems and to pH 8.0 for HMDA activated syste...
