S. cerevisiae BSP-1, BSP-2, BSP-3 and BSP-4 are industrially used baking strains that have same phenotypes, but claimed to be different strains by the industry due to variations in their fermentation activities in dough. In the present study, we have analyzed the rDNA sequences, subtelomeric Y’ sequences and retrotransposon Tyl that are highly repeated in the genomic DNA of the yeast S. cerevisiae. As these three types of repeats have different sequence arrangements in different strains of S. cerevisiae, the genomic DNA of the industrially used strains utilized in this study were subject to a series of cleavage reactions by five different restriction endonucleases (Eco RI, Hind III , Bam HI, Bgl II, Xho I) to interpret the sequence arrangements in each strain. The rDNA sequence in each strain showed no restriction site polymorphism for the enzymes Eco RI, Hind III and Bgl II. The retrotransposon Ty1, when cut by Xho I, generated a same size fragment in all strains and the Y’ subtelomeric sequences also did not indicate any different arrangement when digested with Hind III, Bam HI, and Xho I. The data obtained from this genomic analysis did not support the claims that these strains were different due to variations in fermentation activities in dough, although they have same phenotypes.

Effect of operating parameters on the separation of sugars by nanofiltration
Aydogan, N; Gurkan, T; Yılmaz, Levent (Informa UK Limited, 1998-01-01)
Due to the complexity of fermentation mixtures, separation of solutes and secondary substrates has a special importance. Membrane processes such as nanofiltration may offer good alternatives for the separation of fermentation products or recovery of substrates. For an efficient separation, the membrane type and operating parameters such as feed flow rate, operating pressure, and feed should be optimized. In this study the separation and recovery of sugars were targeted. It was found that with an increase of...
A Statistical Optimization Study on Dilute Sulfuric Acid Pretreatment of Distillers Dried Grains with Solubles (DDGS) As a Potential Feedstock for Fermentation Applications
Çekmecelioğlu, Deniz (2019-11-01)
Distillers dried grains with solubles (DDGS) are co-product of dry-grind ethanol plant, which can be used as a feedstock for fermentation, e.g. for biofuels or other value-added products. However, DDGS loading is a critical factor in the pretreatment, hydrolysis, and fermentation, where the low DDGS loading lowers sugar concentration in the hydrolysate and yields low product concentrations, which can result in high energy cost at the recovery step, while too high loading may have inhibitory effects on the m...
Reassessment of correlations for the dewatering characteristics of filter cakes
Hoşten, Çetin; San, O (2002-05-01)
This paper presents a fundamental study on dewatering characteristics of filter cakes. Laboratory-scale batch pressure-filtration experiments were conducted on quartz, dolomite and andesite samples. having Gates-Gaudin-Schuhmann type of size distributions with different distribution parameters. The residual cake moisture and cake dewatering kinetics data were collected for filter cakes dewatered at varying applied pressure. Correlations for the entry pressure, residual saturation, pore size distribution ind...
Comparative analysis of product and by-product distributions in defined and complex media in serine alkaline protease production by recombinant Basillus subtilis
Oktar, Ceren; Çalık, Pınar; Department of Chemical Engineering (2003)
In this study, firstly the effects of aspartic acid group amino acids -which were reported to be the potential bottleneck in serine alkaline protease (SAP) synthesis- on SAP production were investigated by substituting at a concentration range of 0-15 mM by using recombinant Bacillus subtilis carrying pHV1434::subC gene. All aspartic acid group amino acids except threonine inhibited SAP activity when CAA= 2.5 mM. The highest SAP activities with asparagine, aspartic acid, lysine, threonine, isoleucine and me...
