Site Specific Mutations of Highly Conserved Residues in APH 3 II Phenotypic and Structural Analysis of Mutant Enzymes
Kocabıyık, Semra (null; 1992-05-26)
Site - Specific Mutations of Conserved C - Terminal Residues in Aminoglycoside 3'-Phosphotransferase II [APH(3')-II]: Phenotypic and Structural Analysis of the Mutant Enzymes
Kocabıyık, Semra (1992-06-01)
In order to test the biological importance of amino acids in the C-terminal quarter of aminoglycoside 3′-phosphotransferase II enzyme, seven of the highly conserved residues in this region, His-188, Asp-190, Asp-208, Gly-210, Arg-211, Asp-216 and Asp-220, were changed via site-directed mutagenesis. The phenotype of each mutant was compared to wildtype in terms of antibiotic susceptibilities and enzymatic activities. All of the substitutions either abolished or significantly reduced the resistance of the res...
Site directed mutations of one of the plant pathogen effector gene for functional analysis
Mustafa, Zemran; Akkaya, Mahinur S.; Department of Biochemistry (2015)
Wheat (Triticum aestivum L.) is one of the most important crops planted in the world and used as major food resource. Yet, there are major losses of productivity due to different biotic and abiotic destructive agents. Among the biotic factors especially the yellow rust is one of the most frequent. Yellow rust or also called “stripe rust”, due to stripe like appearances on the leaves, disease is caused by Puccinia striiformis f. sp. tritici, which is an obligate plant fungal pathogen. To cope with high yield...
Sequence specific inhibition of human chemokine gene, cxcl7, by small interfering RNAS in lewis lung carcinoma cells
Kızışar, Dilay; Akkaya, Mahinur S.; Department of Chemistry (2013)
In this study, we aimed to design small interfering RNAs to silence the expression of a gene that is a member of chemokines playing crucial roles on cancer. Small interfering RNAs (siRNAs) are generated by ribonuclease III cleavage of 20-25nt in length dsRNAs which mediate RNAi-induced specific mRNA degradation and they are known to be an important part of the gene regulation mechanism in all Eukaryotes. Recently, siRNAs are being utilized as promising treatment approaches. In this study, we targeted a gene...
Structure Function Analysis for Aminoglycoside 3 Phosphotransferase II APH 3 II
Kocabıyık, Semra (1992-02-01)
Mutant strains containing APH(3')-II were constructed via site-directed mutagenesis of the cloned gene and by random mutagenesis of a strain containing the APH(3')-II gene on a conjugative plasmid. Substitutions at highly conserved amino acid residues produced APH(3') enzymes which in general showed reduced activity and conferred reduced levels of resistance to their substrates. Substitutions at Tyr 218 altered substrate specificity for the enzymes. Random mutagenesis produced plasmid-borne mutations confer...
