Cell cycle-independent furrowing triggered by phosphomimetic mutations of the INCENP STD motif requires Plk1
Papini, Diana; Fant, Xavier; Ogawa, Hiromi; Desban, Nathalie; Samejima, Kumiko; Feizbakhsh, Omid; Askin, Bilge; Ly, Tony; Earnshaw, William C.; Ruchaud, Sandrine (The Company of Biologists, 2019-10-10)
Timely and precise control of Aurora B kinase, the chromosomal passenger complex (CPC) catalytic subunit, is essential for accurate chromosome segregation and cytokinesis. Post-translational modifications of CPC subunits are directly involved in controlling Aurora B activity. Here, we identified a highly conserved acidic STD-rich motif of INCENP that is phosphorylated during mitosis in vivo and by Plk1 in vitro and is involved in controlling Aurora B activity. By using an INCENP conditional-knockout cell li...
CELL CYCLE-DEPENDENT REGULATION OF CXXC5 SYNTHESIS
Demiralay, Öykü Deniz; Muyan, Mesut; Department of Biology (2022-8-22)
17β-estradiol (E2) is the main estrogen in circulation and has many physiological effects on various tissues, including the mammary tissue. CXXC5 is an estrogen-responsive gene product that binds to nonmethylated CpG dinucleotides on DNA. CXXC5 synthesis shows fluctuation in the cell cycle. This led to our prediction that the level of CXXC5 synthesis is regulated through the cell cycle. To test this prediction, I investigated the synthesis of CXXC5 in cell cycle-synchronized cells for every 6h up to 36h. I ...
Cell-surface interactions in a breast cancer model
Antmen, Ezgi; Hasırcı, Vasıf Nejat; Demirci, Utkan; Department of Biotechnology (2017)
Breast cancer, is one of the most commonly diagnosed cancers, has a high mortality rate. One in every eight women (12.3%) develops breast cancer at some stage of their lives and this is the cause of about 15% of cancer deaths in women and 3% of total deaths. It is therefore important to study the behavior of breast cancer cells. Measurement of the mechanical properties of cancer cells leads to new insights such as that cancer cells are softer than healthy cells. Also, metastatic cancer cells were found to b...
Cell behavior on the alginate-coated PLLA/PLGA scaffolds
Bahcecioglu, Gokhan; Hasırcı, Nesrin; Hasırcı, Vasıf Nejat (2019-03-01)
Here, we investigated the effect of preparation temperature and alginate-coating on L929 fibroblast behavior on lyophilized microporous PLIA/PLGA (95:5, w/w) scaffolds. The lower freezing temperature used during lyophilization (-80 degrees C) resulted in smaller pores (around 50 mu m) and higher compressive modulus (1500 kPa) than those prepared at the higher temperature (-20 degrees C) (pore size: 120 mu m, compressive modulus: 600 kPa) (p < 0.01). Cell proliferation was significantly lower on the alginate...
