Effects of metabolic engineering of ethanol utilization pathway in Pichia pastoris on recombinant protein production and transcription levels in central metabolism
Dayankaç , İdil; Çalık, Pınar; Özdamar, Tunçer H.; Department of Chemical Engineering (2021-9)
The aim of this MSc thesis is to increase the ethanol uptake rate and decrease by-product formation in the ethanol utilization (EUT) pathway and acquire a smoothly operating intracellular reaction network in the target microorganism. A metabolically engineered novel host, Pichia pastoris, was developed by overexpression of the two EUT pathway enzymes, alcohol dehydrogenase 2 (ADH2) and Acetyl-CoA synthetase (ACS1) that catalyze rate-limiting reactions, one by one and together, to understand how they affect ...

Effects of ammonium ion, acetate and aerobic conditions on hydrogen production and expression levels of nitrogenase genes in Rhodobacter sphaeroides OU001
Akkose, Sevilay; Gündüz, Ufuk; Yucel, Meral; Eroglu, Inci (2009-11-01)
In the present study, expression levels of nitrogenase encoding nifH and control genes nifA and prrA were examined at different physiological conditions in Rhodobacter sphaeroides O.U.001. In addition to variations in expression levels, changes in hydrogen production and growth were also investigated in response to different concentrations of ammonium source, acetate and aerobic conditions.
Effects of doxorubicin on telomerase and apoptosis in doxorubicin resistant and sensitive MCF-7 cells

Effect of doxorubicin on telomerase activity and apoptotic gene expression in doxorubicin-resistant and -sensitive MCF-7 cells
Eskiocak, Ugur; Iseri, Ozlem Darcansoy; Kars, Meltem Demirel; Bicer, Atilla; Gündüz, Ufuk (2008-01-01)
Background: Dose-and time-dependent effects of doxorubicin on telomerase activity ( TA) and expression levels of hTERT, Bcl-2, Bcl-x(L) and Bax were investigated in doxorubicin-resistant and -sensitive MCF-7 cells. Methods: Doxorubicin-resistant MCF-7/R was developed from sensitive MCF-7 breast carcinoma cell line and acquired resistance was demonstrated by XTT and mRNA analysis of MDR1 and MRP1 genes. Expression levels were determined by RT-PCR. Newly developed rapid and simple TRAP-silver staining assay w...
