The purpose of the study is to purify human growth hormone from the fermentation broth by affinity chromatography. For this purpose, human growth hormone specific oligonucleotide aptamers are selected among an aptamer library; selected oligonucleotides were synthesized and used as ligands. Effect of pH on ligand-human growth hormone complex formation was investigated and the highest complex formation was obtained at pH= 7.0. Human growth hormone is separated from the fermentation broth with 99.8% purity and 41% overall yield. The equilibrium data obtained was described by Langmuir type isotherm where saturation constant (q0) and affinity constant (K) are calculated as 0.338 mg hGH/mol aptamer and 0.059 mg hGH/ml, respectively. Further, equilibrium data obtained using aptamer affinity column was described by Langmuir type isotherm where saturation constant (q0) and affinity constant (K) are 0.027 mg hGH/mol aptamer and 1.543 mg hGH/ml, respectively. It is possible that, selected aptamer can be used for purification of bulk amounts of recombinant human growth hormone by using aptamer affinity chromatography.

We present an example of expression and purification of a biologically active G-protein coupled receptor (GPCR) from yeast. An expression vector was constructed to encode the Saccharomyces cerevisiae GPCR a-factor receptor (Ste2p, the STE2 gene product) containing a 9-amino acid sequence of rhodopsin that served as an epitope/affinity tag. In the construct, two glycosylation sites and two cysteine residues were removed to aid future structural and functional studies. The receptor was expressed in yeast cell...
Equilibrium studies on the reactive extraction of lactic acid from fermentation broth
Açan, Başak; Gürkan, Türker; Department of Chemical Engineering (2003)
Lactic acid recovery from dilute fermentation broths is a growing requirement due to the increasing demand for pure lactic acid. Reactive extraction is proposed as an alternative to conventional methods of recovery, since the selectivity of separation is remarkably enhanced in reactive extraction. The aim of this study is to perform the equilibrium studies for the recovery of lactic acid from its synthetic aqueous solutions (not from real fermentation broths) by reactive extraction and investigate the effec...
Kinetic analysis of aspergillus oryzae cultivations on starch
Bayındırlı, Alev; UNGAN, S (Informa UK Limited, 1991-01-01)
Cultivation of Aspergillus oryzae on starch is described as a combination of two rate processes: Starch hydrolysis and the cellular activities of the fungi including growth, enzyme production and maintenance. Kinetic models are presented to describe growth, enzyme production, starch hydrolysis and uptake of the hydrolysis products. Numerical values of the model parameters indicated that the rate controlling step of A. oryzae growth on starch was not starch hydrolysis, but the substrate uptake process. Gluco...
Estimation of lipid properties related to supercritical fluid extraction
Yener, Meryem Esra (2001-01-01)
The correlations for vapor pressures of fatty acids, fatty acid methyl and ethyl esters, and simple triglycerides were given based on the available data in the literature. The developed equations enable easy estimation of the critical properties. The group contribution methods were used to develop the estimation method. The estimated properties of the most common lipids were tabulated and the differences between the estimation methods were critically discussed.
Optimization of dilute sulfuric acid, aqueous ammonia, and steam explosion as the pretreatments steps for distillers' dried grains with solubles as a potential fermentation feedstock
Iram, Attia; Çekmecelioğlu, Deniz; Demirci, Ali (Elsevier BV, 2019-06-01)
Distillers' dried grains with solubles (DDGS) is the by-product of bioethanol production from starch-rich grains through dry-mill fermentation. In this study, dilute sulfuric acid hydrolysis, aqueous ammonia, and steam explosion as the pre-treatment methods were optimized. The central composite response surface methodology (RSM) design was used for optimization of dilute acid pretreatment, aqueous ammonia pretreatment. The steam explosion trials were evaluated. The results show that the dilute acid pretreat...
