Osteomyelitis is a difficult to treat bone infection. In clinic, osteomyelitis is treated with surgical debridement followed by the antibiotic therapy. But, due to the low blood circulation in the infection area and the production of an exopolysaccharide, biofilm, by the pathogen violence the treatment. Therefore local drug delivery systems are generated. These systems transmit the effective dose to the infection area without any toxic effects. In this study, vancomycin containing PDLLA/β-TCP and PLGA/β-TCP composites were developed and characterized. Vancomycin containing PDLLA/β-TCP and vancomycin containing PLGA/β-TCP composites released 3.1 ± 0.2 mg and 3.4 ±0.4 mg vancomycin for six weeks, respectively and the released vancomycin was sufficient for MRSA susceptibility. Also, the released vancomycin inhibited the biofilm formation of Methicillin Resistant S. aureus. Vancomycin containing PDLLA/β-TCP and vancomycin containing PLGA/β-TCP composites were biocompatible with Mesenchymal Stem Cells and SaOS-2 osteosarcoma cells. The composites showed alkaline phosphatase activity at day 21. But for the RT-PCR studies conducted with the extraction medium of the composites, the vancomycin containing PDLLA/β-TCP and vancomycin containing PLGA/β-TCP composites were not induced the differentiation of mesenchymal stem cells by not expressing some of the osteogenesis related signalling molecules.

Vancomycin containing plla delivery system for bone tissue biocompatibility and treatment of implant related chronic osteomyelitis
Uysal, Berna; Korkusuz, Feza; Department of Biotechnology (2009)
Osteomyelitis is an infection of bone or bone marrow, usually caused by pyogenic bacteria. It can cultivate by hematogen way or it can cultivate by the help of local soft tissue infection. Osteomyelitis often requires prolonged antibiotic therapy and surgery. But for therapy; antibiotic must reach to effective dose in the bone. So that; for prevention and treatment of osteomyelitis controlled antibiotic release systems can be used. These systems have been developed to deliver antibiotics directly to infecte...
In vitro and in vivo evaluation of doxycycline-chondroitin sulfate/PCLmicrospheres for intraarticular treatment of osteoarthritis
AYDIN, Ozlem; KORKUSUZ, FEZA; KORKUSUZ, PETEK; Tezcaner, Ayşen; BİLGİÇ, ELİF; Yaprakci, Volkan; Keskin, Dilek (2015-08-01)
Osteoarthritis (OA) is a degenerative joint disease, which has no complete treatment with medication yet. Intraarticular hyaluronan (HA) injection can decrease pain and modify the natural course of OA. This study was designed to provide long term delivery of an MMP (matrix-metalloproteinase) inhibitor agent-doxycycline, together with matrix regenerative agent-chondroitin sulfate for treating OA which progress with matrix degenerations. Doxycycline (D) and doxycycline-chondroitin sulfate (D-CS) loaded poly--...
Numerical Study on Blood Flow Modelling in Arteries
Kaya Merdan, Songül (null; 2018-06-29)
In order to prevent, diagnose and treat vascular disease, detailed knowledge of blood blow and the response of blood vessels is essential. The use of computer simulations can provide researchers extremely useful tool of the interpretation and analysis of the circulatory sysytem in both physiological and pathological situations. In this work, we present a 2D mathematical model to forecast for some cardivascular diseases such as stenosis and aneurysms geometry. We consider a blood flow based on Stokes and Nav...
Biological Responses of Ceramic Bone Spacers Produced by Green Processing of Additively Manufactured Thin Meshes
Minguella-Canela, Joaquim; Calero, Jose Antonio; Korkusuz, Feza; Korkusuz, Petek; Kankılıç, Berna; Bilgiç, Elif; De los Santos-López, M. Antonia (MDPI AG, 2020-5-30)
Bone spacers are exclusively used for replacing the tissue after trauma and/or diseases. Ceramic materials bring positive opportunities to enhance greater osteointegration and performance of implants, yet processing of porous geometries can be challenging. Additive Manufacturing (AM) opens opportunities to grade porosity levels in a part; however, its productivity may be low due to its batch processing approach. The paper studies the biological responses yielded by hydroxyapatite with beta -TCP (tricalcium ...
Investigation Of The Structure Of Celecoxib Molecule And Its Interaction With Cox-2 By Quantum Mechanical Methods Using Model Systems
Kıran, Aleyna Dilan; Selçuki , Cenk (Orta Doğu Teknik Üniversitesi Enformatik Enstitüsü; 2022-10)
Non-steroidal anti-inflammatory drugs (NSAID) inhibit the eventual conversion of COXs to metabolites of arachidonic acid, including prostaglandins, prostacyclin, and thromboxanes, in the immune response after injury or infection. Selective COX-2 inhibitors (coxibs), a subset of NSAIDs, act specifically on the COX-2 enzyme. One of the coxibs, celecoxib, is marketed under the brand name Celebrex. Celebrex is a non-steroidal anti-inflammatory drug that exhibits anti-inflammatory, analgesic and antipyretic acti...
