Colorectal cancer is a disease resulting from the malignant transformation of colon epithelia. As of 2013, it the third leading cause of death among cancer related deaths, in Turkey. Lipoxygenases (LOXs) are a family of fatty acid dioxygenases that convert polyunsaturated fatty acids (PUFAs) to their oxidized form, which are subsequently transformed to bioactive lipid mediators acting in various cell functions. Specifically, 15-Lipoxygenase-1 (15-LOX-1) is a member of the LOX family, oxygenating linoleic acid (LA) preferentially to 13-(S)-HODE as the final product. 15-LOX-1 is known to be important in the resolution of inflammation; the enzyme also has tumor suppressive properties and it can get downregulated in several different cancer types. 15-LOX-1 re-expression in colorectal cancer was shown to decrease metastasis, cell proliferation and invasive capabilities of the cells. We wanted to examine whether re-expression of the enzyme would also lead to a decrease in angiogenic properties. In this context, we assayed the VEGF transcript and secreted protein levels and treated HUVEC cells with conditioned media from SW480 cells expressing 15-LOX-1 . We observed a decrease in both VEGF transcript levels and secreted protein levels in both normoxic and hypoxic conditions. Additionally, HUVECs incubated with conditioned media collected from 15-LOX-1 transfected SW480 cells showed reduced formation of tube-like structures in vitro. In conclusion, 15-LOX-1 re-expression was shown to reduce angiogenic abilities of the colorectal cancer cells.

Effect of gemcitabine and retinoic acid loaded PAMAM dendrimer-coated magnetic nanoparticles on pancreatic cancer and stellate cell lines

Gemcitabine is an anticancer drug used in the treatment of different cancer types, including pancreatic ductal adenocarcinoma. The maximum tolerated dose in humans is restricted by its side effects on healty cells. Furthermore, the fibrotic stroma produced by the pancreatic stellate cells prevents effective delivery of chemotherapeutic agents providing a safe-haven for the cancer cells. This becomes more of a problem considering the short half-life of this drug. Magnetic nanoparticle-based targeted drug del...
Investigation of the effects of plant phenolic oleuropein on expression levels of xenobiotic metabolizing enzymes along with potential cytotoxic and genotoxic impact on human colon cancer cell line HT-29

Colon cancer is one of the major health problems causing malignancies and is frequently encountered in developed western countries. Today, in the treatment of colon cancer, several methods are used, mostly with adverse effects. In order to reduce the consequences of side effects usage of natural or artificial anti-cancer molecules is a serious topic in the cancer research area. Scientists began to perform research to find anti-carcinogenic phytochemicals and xenobiotics with anti-carcinogenic potentials. Un...
Effect of MDR modulators verapamil and promethazine on gene expression levels of MDR1 and MRP1 in doxorubicin-resistant MCF-7 cells

One of the major problems of cancer chemotherapy is the development of multidrug resistance (MDR) phenotype. Among the numerous mechanisms of MDR, a prominent one is the increased expression of membrane transporter proteins, the action of which leads to decreased intracellular drug concentration and cytotoxicity of drugs. Among them, P-gp and MRP1, encoded by MDR1 and MRP1 genes, respectively, have been associated with MDR phenotype. Chemical modulators can be used to reverse MDR. These chemicals can either...
Effect of cobalt-60 (gamma radiation) on multidrug-resistant multiple myeloma cell lines

Emergence of resistance to chemotherapy and radiotherapy is a major obstacle for the successful treatment of MM (multiple myeloma). Prednisone, vincristine and melphalan are commonly used chemotherapeutic agents for the treatment of MM. In the current study, we examined the presence of possible cross-resistance between these drugs and gamma (gamma) radiation. Prednisone, vincristine and melphalan resistant RPMI-8226 and U-266 MM cells were generated by stepwise increasing concentrations of the drugs. The se...
Effects of novel compound derived from vitamin e on prostate cancer cell line

Cancer is a complex disease characterized by uncontrolled cell proliferation, invasion and metastasis. It is known that vitamins having high antioxidant capacity, like vitamin A, C and E, play important roles in adjuvant therapy and as cancer protective agents. Vitamin E is a group of fat soluble antioxidant nutrients consisting of tocopherols and tocotrienols. Various research has been performed on the utility of vitamin E derivatives on cancer treatment. Energy metabolism of cells alters during cancer pro...
