In the present study antileukemic enzyme L-asparaginase (ASNase) and catalase (as a model enzyme) were modified in solid-phase with activated polyethylene glycol (PEG,) by using ligand-immobilized affinity column systems L-asparagine-Sepharose CL-4B and Procion red-Sepharose CL-4B, respectively. Studies on change of specific activity with modification time showed negligible differences between batches of modified catalase. Modification of ASNase for I It resulted in 50.2% recovery of the specific activity and the attachment of 69 molecules of PEG(2) per molecule of ASNase forming 'PEGylated ASNase'. Sequential modification of ASNase by activated PEG and heparin resulted in coupling of about nine molecules of heparin per molecule of PEGylated ASNase. Intravenous (i.v.) administration of PEG(2)-modified ASNase showed prolonged presence in the blood circulation and no adverse effects or symptoms of anaphylaxis were observed in presensitized mice.

MATRIX ENTRAPMENT OF GLUCOSE-OXIDASE BY GAMMA-IRRADIATION
GURSEL, I; Hasırcı, Vasıf Nejat (1992-01-01)
In this study, matrix entrapment of the enzyme glucose oxidase was achieved through gamma-irradiation of monomers N-vinyl pyrrolidone, 2-hydroxyethyl methacrylate and their mixture. To test the effect of radiation on entrapment efficiency, retention of activities and properties of the system, duration and temperature were varied. The reusability of the resultant products was tested. It was generally found that inclusion of the hydrophilic monomer N-vinyl pyrrolidone into the matrix increased the water conte...
Removal of cobalt from zinc sulfate solution by cementation prior to zinc electrowinning
Kayın, Pınar Burcu; Topkaya, Yavuz Ali; Department of Metallurgical and Materials Engineering (2003)
The aim of this study was to investigate the removal of cobalt from zinc sulfate solution by cementation with the help of conventional and new type of additives that were 4% Sn-zinc alloy powder and 10% Sn-zinc alloy powder, respectively. Synthetic leach solutions containing 150 g/l Zn and 75 mg/l Co were prepared and used in all of the experiments. The parameters researched with the conventional method were the amount of arsenic trioxide and the effect of copper sulfate on cementation of cobalt. While usin...
Benzoylformate decarboxylase from Pseudomonas putida as stable catalyst for the synthesis of chiral 2-hydroxy ketones
Iding, H; Dunnwald, T; Greiner, L; Liese, A; Muller, M; Siegert, P; Grotzinger, J; Demir, Ayhan Sıtkı; Pohl, M (2000-04-14)
The thiamin diphosphate- and Mg2+-dependent enzyme benzoylformate decarboxylase (BFD) from Pseudomonas putida was characterized with respect to its suitability to catalyze the formation of chiral 2-hydroxy ketones in a benzoin-condensation type reaction. Carboligation constitutes a side reaction of BFD, whereas the predominant physiological task of the enzyme is the non-oxidative decarboxylation of benzoylformate. For this purpose the enzyme was obtained in sufficient purity from Pseudomonas putida cells in...
Positive Matrix Factorization dynamics in fingerprinting: A comparative study of PMF2 and EPA-PMF3 for source apportionment of sediment polychlorinated biphenyls
Karakas, Filiz; İmamoğlu, İpek; Gedik, Kadir (2017-01-01)
Receptor models were typically used in air pollution studies and few publications are available for Positive Matrix Factorization (PMF) that consider the details of parameters and procedures in evaluating the trace organic pollutants in sediments. In this study, environmental fate and source composition of Lake Eymir sediments contaminated by polychlorinated biphenyls (PCBs) were explored by applying two PMF models, Paatero's PMF2 and United States Environmental Protection Agency's (US EPA) EPA-PMF3. PMF2 a...
Enzyme electrodes fabricated by dad type poly (2,5-di(furan-2-yl)thiazolo[5,4-d]thiazole) conducting polymer
Kurzama, Nadia; Söylemez, Saniye; Böyükbayram, Ayşe Elif; Toppare, Levent Kamil (2020-03-20)
Poly(2,5-di(furan-2-yl)thiazolo[5,4-d]thiazol) polymer was coated over a platinum electrode and new enzyme electrodeswere produced that are not found in the literature. The formation of polymerization was shown by cyclic voltammetry.Kinetic parameters of immobilized polyphenol oxidase enzyme were determined. Vmax and Km were obtained as 0.028 ±0.001 umol min1- electrode1- and 669.68 ± 64.73 mM respectively. The effect of reaction conditions over enzyme activitywere examined. The pH and temperature at which ...
